摘  要:目的研究新乡地区固定单采血小板献血者HPA1-18系统基因多态性分布,为输血患者寻找相容性血小板提供可靠依据。方法采用PCR-SSP法对150名固定单采血小板献血者标本进行HPA1-18系统的基因分型。结果HPA1-18系统基因在本地区固定献血者中分布频率分别为HPA-1a 0.990和-1b 0.010,HPA-2a 0.947和-2b 0.053,HPA-3a 0.376和-3b 0.624,HPA-15a 0.476和-15b 0.524;其余HPA系统只检测到a等位基因,故其基因频率均为1.000。结论HPA-2、3、15系统杂合度较高,抗原分布不配合率相对高,在临床配合性血小板输注中必须加以重视。Objective To investigate the allele frequencies of the HPA 1-18 system in Xinxiang regular apheresis platelet donors, so as to provide the credible basis for screening compatible platelets for patients receiving transfusion. Methods The genotypes of HPA 1-18 system were determined by polymerase chain reaction using sequence-specific primer (PCR- SSP) on 150 samples. Results Gene frequencies were 0.990 and 0.010 for HPA-1 a and -1 b, 0.947 and 0.053 for HPA- 2a and -2b, 0.376 and 0.624 for HPA-3a and -3b, and 0.476 and 0. 524 for HPA-15a and -15b. All the individuals tested were homozygotes for HPA-4a-14a and HPA-16a-18a. There was a good fit to Hardy-Weinberg equilibrium within each group. Conclusion The highest possibility of heterozygotes occurs in HPA-3 and HPA-15, which incompatible frequency of HPA antigen distribution is the highest. More attention must be paid to its role in platelet match for clinical transfusion.
