摘  要:目的优选三七总皂苷提取和大孔吸附树脂纯化工艺。方法以三七皂苷R1、人参皂苷Rg1和人参皂苷Rb1含量为指标,以乙醇浓度、乙醇用量、提取时间、提取次数为考察因素,采用正交试验优选提取工艺;以比吸附量和洗脱率为指标,采用大孔吸附树脂纯化三七总皂苷。结果最佳提取工艺为:体积分数50%乙醇,回流提取2次(10倍、8倍量),每次2.0 h;纯化工艺条件为:选用HPD-100型大孔吸附树脂,调节上样溶液浓度约17 mg/m L,最大上样体积为3.3 BV,流速1.0 m L/min,80%乙醇4 BV洗脱,洗脱流速2.0 m L/min;三七总皂苷平均得率10.18%,其中三七皂苷Rl,人参皂苷Rgl和Rbl的总含量大于65%。结论优选的提取、纯化工艺稳定,三七皂苷类成分转移率高,工艺简便,适合工业化生产。Objective To optimize the extraction process and establish a purification method for total saponins in Notoginseng radix et rhizoma with macroporous resin.MethodsWith the contents of notoginsenoside R1, ginsenoside Rg1 and ginsenoside Rb1 as indices, the extraction technology ofNotoginseng radix et rhizoma was optimized by orthogonal design with the concentration and amount of ethanol and extraction time and times as factors. The macroporous adsorption resin was used to purify total saponins with specific adsorption capacity and elution rate as indices.Results The optimum extraction conditions were as follows: extracted 2 times with 10- and 8-fold 50 % ethanol, 2 h each time. The purification conditions were as follows: HPD-100 macroporous adsorption resin was chosen, the concentration of total saponins sample solution was about 17 mg/mL, the maximum sample volume was 3.3 BV, with the flow rate of 1.0 mL/min, 4 BV 80 % alcohol was used as eluant with the flow rate of 2.0 mL/min. The yield of total saponins was 10.18 %, and the contents of notoginsenoside R1, ginsenoside Rgl and ginsenoside Rblexceeded 65 %. Conclusion This optimized extraction and purification process is convenient and stable, also with high transfer rate of total saponins, and can be used for the industrial production.
