摘 要:目的探讨藏方五味余甘子散对大鼠心肌缺血再灌损伤相关细胞凋亡蛋白表达的影响及其可能机理。方法 96只SD大鼠随机分成6组,分别为五味余甘子散高、中、低剂量组,伪手术组,模型组以及阳性对照组,每组16只。用结扎大鼠左冠状动脉前降支法制备心肌缺血再灌注模型,再灌注40 min后,检测大鼠心肌细胞Bcl-2、Bax蛋白的表达。结果五味余甘子散能显著提高心肌细胞Bcl-2蛋白的表达,抑制Bax蛋白的表达,同时提高Bcl-2/Bax比值。结论五味余甘子散对大鼠心肌缺血再灌损伤具有保护作用,其机制可能通过上调凋亡Bcl-2蛋白表达,提高Bcl-2/Bax比值,下调Bax蛋白的表达抑制细胞凋亡。Objective To explore the influence of apoptosis - associated protein expression and its possible mechanism of Wuwei Yuganzi San, one of Tibetan medicine, on myocardial ischemia - reperfusion injury in rats. Methods Ninety - six Sprague - Dawley rats were assigned randomly into six groups including high dose group, moderate dose group and low dose group according to the dosage of Wuwei Yuganzi San administrated. Moreover, Sham -operation group, Model group and Positive control group were supplemented. And each group was 16 rats, Myocardial ischemia -reperfusion injury model was established by ligation of the left anterior descending coronary artery (LAD) of the heart. Following 40 min reperfusion in rats, Bcl - 2 and Bax protein expression in cardiomyocytes were detected. Results Myocardial Bcl - 2 protein expression was significantly augmented well paralleling with an increase in ratio of Bcl -2 to Bax, while Bax protein expression was inhabited. Conclusion WuweiYuganzi San may have a protective effect on myocardial ischemia - reperfusion injury model. The mechanisms may associate with an up - regulated expression of Bcl - 2, and an increased ratio of Bcl - 2/Bax and a low - regulated expression of Bax, resulting in suppression of apoptosis.
