番禺地区A型人群中A2亚型的初探

摘　　要：目的调查分析番禺地区A型人群中A2亚型的分布频率和基因分型的特征。方法使用抗-A1试剂(植物凝集素),对经单克隆ABO正定型试剂确定为A型的样本进行96孔微量板法快速筛查;筛查阴性的样本采用人源抗-A1试剂进行试管法血清学鉴定;采用PCR-SSP法(A2基因分型检测试剂盒)对抗-A1阴性样本的基因组DNA进行扩增,对扩增产物进行电泳分析,根据电泳图谱判断A2基因分型。结果在1 656例A型样本中筛查和鉴定检出5例抗-A1阴性,发生频率约为0.3%。基因分型:2例A201,2例A205,1例为不确定,需进一步测序分析。结论本地区A型人群中A2亚型发生频率约为0.3%;A2亚型中存在A201和A205等基因型;使用血清学结合PCR-SSP方法能快速鉴定A2亚型。Objective To analyze the frequency and genotype characteristics of A2 blood subtype in type A population of Panyu,Guangdong province.Methods Anti-A1 phytagglutinin was used to quickly screen A blood group samples,The A blood group samples that had been identified by monoclonal ABO positive stereotypes reagents were screened using human anti-A1 reagent and PCR-SSP（A2gene variants genotyping assay kit to amplify template DNA of anti-A1 negative samples）,followed by electrophoresis of the PCR products to determine A2 genotype.Results Five anti-A1 negative samples were obtained from 1 656 samples of A blood group,the negative frequency was 0.3%.Genotyping revealed 2cases of A201,2cases of A205,and one case which needs further sequencing analysis.Conclusion In Panyu region,the frequency of A2 subtype in the A blood group population is approximately 0.3%.The A2 subtype includes A2 01,A2 05 and other genotypes.Serology method combined with PCR-SSP may quickly identify A2 subtypes of A blood group.
