摘 要:目的探讨过氧化氢(H_2O_2)通过高迁移率族蛋白(HMGB1)介导对心肌损伤的作用机制。方法胎鼠心肌细胞培养后,分为4组:对照组,H_2O_2不同浓度组(100μmol/L、200μmol/L、500μmol/L),H_2O_2(200μmol/L)+乙酰半胱氨酸(NAC,200μmol/L)组,H_2O_2(200μmol/L)+HMGB1抗体(20μg/ml)组。检测心肌细胞活性、凋亡率、乳酸脱氢酶(LDH)、肌酸激酶(CK)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)、HMGB1表达。结果(1)细胞活性:与对照组比较,心肌细胞活性随着H_2O_2浓度的增加而降低(t=-5.81,-9.59,-14.21,P均<0.05);NAC和HMGB1抗体可以明显减弱H_2O_2(200μmol/L)对细胞活性的抑制作用(t=-2.98,-3.03,P<均0.05)。(2)LDH和CK的表达水平:与对照组比较,LDH和CK的表达量随着H_2O_2的浓度增加而增加(t_(LDH)=9.78,15.37,18.13,t_(CK)=11.29,20.35,26.17,P均<0.05)。与H_2O_2(200μmol/L)组比,NAC和HMGB1抗体可以抑制LDH和CK的表达(t_(LDH)=-4.91,-4.22,t_(CK)=-4.17,3.29,P均<0.05)。(3)MDA表达和SOD活性:H_2O_2(100,200,500μmol/L)可以抑制心肌细胞SOD活性,增加MDA表达(t_(SOD)=-4.16,-6.59,-10.76,t_(MDA)=3.98,5.16,9.47,P均<0.05);与H_2O_2(200μmol/L)组比,NAC和HMGB1抗体可以拮抗H_2O_2对SOD和MDA的影响(t_(LDH)=-4.91,-4.22,t_(CK)=-4.17,3.29,P均<0.05)。(4)HMGB1的表达:与对照组比较,H_2O_2(100,200,500μmoL/L)可明显增加心肌中HMGB1的表达(t=12.94,21.08,25.81,P均<0.05),与H_2O_2(200μmol/L)组比,NAC+H_2O_2和HMGB1-Ab+H_2O_2中HMGB1的表达明显降低(t=-10.24,-8.09,P均<0.05)。(5)心肌凋亡:与对照组比较,H_2O_2组(100、200、500μmol/L)心肌凋亡率明显增加(t=7.52,13.06,16.61,P<0.05);NAC+H_2O_2组和HMGB1+H_2O_2组心肌凋亡率较H_2O_2(200μmol/L)组明显增加(t=-4.89,-4.23,P<0.05)。结论 H_2O_2诱导的氧化应激反应可以通过上调HMGB1表达,损伤心肌。O bjective To investigate the effect of hydrogen peroxide ( H2 O2 ) by high mobility group protein ( HMGB1) mediated mechanism of myocardial injury.Methods Fetal rat myocardial cells cultured and divided into 4 groups:control group, H2 O2 different concentration group (100μmol /L, 200 μmol /L, 500μmol /L), H2O2(200μmol/L) +N-acetylcysteine (NAC, 200μmol /L ) group, H2O2(200μmol /L) +HMGB1 antibody (20 ug /ml) group.The activity of myocardial cells, apoptosis rate, lactate dehydrogenase ( LDH) , creatine kinase( CK ) , superoxide dismutase (SOD)activityandmalondialdehyde(MDA)andHMGB1expressionweredetected.Results (1)Cellactivity:compared with the control group , myocardial cell activity along with the increase of H2 O2 concentration were decrease ( t =-5.81, t =9.59, t =-14.21, all P 〈0.05);NAC and HMGB1 antibodies can be significantly reduced H2 O2 (200 umol /L) on the&amp;nbsp;activity of cell inhibition ( t =-2.98, t =-3.03, P 〈0.05) .(2) The expression level of LDH and CK:compared with the control group, the expression of LDH and CK were increased along with the increase of the H2 O2 concentration ( LDH, t =9.78, t =15.37, t =18.13.CK, t=11 .29, t =20.35, t =26.17, all P 〈0.05).Compared with H2O2(200umol /L) group, NAC and HMGB1 antibodies can inhibit LDH and CK expression (LDH, t =-49.1 , t =-4.22, CK, t =-4.17, t =3.29, all P 〈0.05).(3) The expression of MDA and SOD activity:H2O2(100, 200, 500 umol /L) can inhibit the ac-tivity of SOD of myocardial cells, increase the expression of MDA (SOD, t =-4.16, t =-6.59, t =-10.76, MDA, t =3.98, t =5.16, t =9.47, P 〈0.05);H2 O2 (200 umol /L) group, NAC and HMGB1 antibodies can antagonize the H2 O2 on superoxide dismutase (SOD) and malondialdehyde (MDA) (LDH, t =-4.91, t =-4.22, CK, t =-4.17, t =3.29, all P 〈0.05).(4) The expression of HMGB1:compared with the control group, H2 O2(100, 200, 500 umol /L) can be significantly in
