逆针灸对去势雌性大鼠自发性乳腺肿瘤的防治作用研究

摘　　要：目的:观察逆针灸对去势雌性大鼠自发性乳腺肿瘤的防治作用,并探讨其作用机制。方法:将6月龄SD雌性大鼠250只随机分为空白组(30只),假手术组(40只),去势模型组(180只)。假手术组切除卵巢附近与卵巢大小相同的脂肪组织一块,造模组完整切除双侧卵巢。造模3个月后,再随机将造模组剩余的大鼠(在造模过程中死亡7只,其他的用于做与本实验相关的实验)分为逆针3月组(40只),逆针6月组(40只)和对照组(40只)。采用电化学发光免疫方法,分别于造模3个月后和治疗结束后从腹腔静脉采血做各组血清肿瘤标志物检测。计数包块数量并记录形态,取包块组织,用精度为1 mm的游标卡尺测量包块的高度、最大直径和相垂直的直径。Olympus光学显微镜下观察包块组织病理学变化,确定包块类型。结果:与逆针3月组相比,空白组,假手术组和对照组乳腺肿瘤发生率明显升高,乳腺肿瘤的数目明显增多,乳腺肿瘤的体积明显增大,差异均有统计学意义(均P<0.05);与逆针6月组相比,空白组,假手术组和对照组乳腺肿瘤发生率明显升高,乳腺肿瘤的数目明显增多,乳腺肿瘤的体积明显增大,差异均有统计学意义(均P<0.05);治疗前血清CA153空白组,假手术组,造模组之间均无显著差异(P=0.782,P=0.886,P=0.843)。组内血清CA153比较,空白组治疗前低于治疗后(P<0.05),逆针组治疗前后无显著差异(P=0.786)。结论:逆针灸能够降低去势雌性大鼠自发性乳腺肿瘤的发病率,发生数目和发生体积,其作用机制可能与调节机体性激素及受体水平及其信号传导,提高机体免疫能力和调控人体基因表达有关;逆针灸有双向调节血清或血浆中糖蛋白CA15-3浓度的作用。Objective ：To observe the inverse acupuncture on castrated female rats spontaneous breast tumor prevention and treatment function and to explore its action mechanism. Methods：6 months old 250 SD female rats were randomly divided into blank group（30）, control group（40）, castrated model group （180）. Control near ovaries removed fat tissue piece with the same size with the ovary, build module complete resection of bilateral ovaries. After 3 months building, random module of the rest of the rat was to be built（ death in the process of building 7 ,the other were used to do this experiment related experiment） march into re- verse needle group （40）, inverse needle June group （g0） and the control group （40）. Using electrochemical luminescence immune method, respectively after 3 months building and treatment the abdominal cavity venous blood was for serum tumor markers detec- tion. Count bag piece number and record form, mass organizations, with accuracy of I mm vernier calipers to measure the height of bag piece, maximum diameter and the diameter of the vertical. Push around optical microscope bag piece histopathological chan- ges, determine the type of bag piece. Results ： Compared with 3 months inverse needle group and blank group, control group and the sham group significantly increased incidence of breast cancer, breast cancer significantly increased, the number of breast tumor volume increased obviously, the difference had statistical significance （P 〈 0.05 ）.
