摘 要:目的:观察"祛风通窍方"对血管性痴呆大鼠海马及额叶皮层神经元细胞变化的影响及作用机制。方法:用Morris水迷宫筛选符合要求SD大鼠60只,随机分为假手术组、模型组、阳性组、祛风通窍方高、中、低剂量组每组各10只。手术组采用2-VO法制备模型,造模1月后,阳性组予尼莫地平6.25 mg/(kg·d)灌胃,祛风通窍方高、中、低剂量组分别给予祛风通窍方26.8 g/(kg·d)、13.4 g/(kg·d)、6.7 g/(kg·d)灌胃,假手术组、模型组予等体积蒸馏水灌胃,连续30 d。心脏灌注内固定,取脑组织,观察各组大鼠海马CA1区锥体细胞及额叶皮层神经元线粒体的变化。结果:光镜下海马CA1区锥体细胞变化:与假手术组比较,模型组锥体细胞丢失明显,细胞1~2层,排列散乱;界限不清,大量细胞变形,核固缩碎裂、呈深蓝色、胞浆浓缩;与模型组比较,尼莫地平组及高、中剂量组锥体细胞层次、排列、形态得到了明显改善。电镜观察额叶皮层神经元线粒体变化:与假手术组比较,模型组线粒体明显水肿破裂,线粒体嵴结构不清晰,线粒体部分固缩;与模型组比较,尼莫地平组,祛风通窍方高、中剂量组线粒体受损情况得到了明显改善。结论:"祛风通窍方"可明显改善血管性痴呆大鼠海马CA1区锥体细胞形态及额叶皮层神经元线粒体的超微结构。Objective:To observe Qufeng Tongqiao Fang effect and mechanism on vascular dementia rat hippocampus and fron- tal cortex neurons change. Methods:Morris water maze screening to meet the requirements,60 SD rats were randomly divided into sham operation group, model group, positive group, Qufi^ng Tongqiao Fang high, medium, low -dose groups, 10 in each group. Surgical group were treated with 2 - VO model prepared by modeling. After operation, positive group was given nimodipine 6.25 rag/( kg· d) gavage, Qufeng Tongqiao Fang groups were given Qufeng Tongqiao Fang 26.8 g/( kg · d), 13.4 g/( kg· d), 6.7 g/( kg · d) orally, sham operation group and model group were given an equal volume of distilled water for 30 days. Cardiac per- fusion fixation and fetch brain were used to observe the changes of each group hippocampus CA1 pyramidal cells and neurons of the prefrontal cortex mitochondria. Results:Light microscope, the changes of hippocampus CA1 pyramidal cells:compared with the sham group, the loss of pyramidal cells of the model group, 1 to 2 layers of ceils arranged in scattered; ill - defined, a large number of cell deformation, nuclear condensation and fragmentation was dark blue, cytoplasm concentration;compared with model group, nimodipine group, high and medium dose group, pyramidal cells arranged in the form significantly improved. The changes of elec- tron microscopy prefrontal cortex neuronal mitochondria:Compared with the sham group and model group, the mitochondria edema rupture of the mitochondrial cristae structure was not clear, some pyknotic mitochondria ; compared with model group, nimodipine group, high and mediun dose groups of Qufeng Tongqiao Fang mitochondria damage significantly improved. Conclusion: Qufeng Tongqiao Fang can significantly improve vascular dementia rat hippocampus CA1 pyramidal cell morphology and frontal cortex neurons in the mitochondrial ultrastructure.
