摘  要:目的 建立测定益母草颗粒中盐酸水苏碱、盐酸益母草碱含量的HPLC测定法.方法 采用高效液相色谱法,以Kromasil 100-5NH2(Dimensions 250×4.6 mm,5μm)E78824和AgilentTC-C18(2)(250×4.6 mm,5μm)588925-902为色谱柱,检测波长:192 nm和277 nm,流动相:乙腈-水(80∶ 20)、乙腈-0.3%庚烷磺酸钠的0.1%磷酸溶液(24:76),流速为1.0 mL/min.理论板数:按盐酸水苏碱峰计算应不低于5000、按盐酸益母草碱峰计算应不低于8000.结果 盐酸水苏碱在0.18~1.08 mg范围内与峰面积呈良好的线性关系,r=0.99998,平均回收率为99.6%,RSD小于2.0%;盐酸益母草碱在7~41μg范围内与峰面积呈良好的线性关系,r=0.99998,平均回收率为99.8%,RSD小于2.0%.结论 该方法操作简便、快速、准确、灵敏度高、重现性好,适用于益母草中盐酸水苏碱、盐酸益母草碱的含量测定.Objective To establish a method for content determine of hydrochloric stachydrine and hydrochloric leo- nurine in motherwort granules. Methods An HPLC method was adopted. The separation was carried out on a Kroma- sil 100- 5NH2( Dimensions 250 ×4.6 mm,5μm) E78824 column and a Agilent TC- C18(2)(4.6 ×250 mm,5 μm) 588925 -902 column. The UV detective wavelength was set at 192 nm and 277 nm. Acetonitrile -water ( 80 : 20) and acetonitrile - 0.3 % heptane sulfonate diluted by 0.1% phosphate (24:76) as mobile phase at a flow rate of 1.0 mL/min. Theoretical plate number were below 5000 and 8000 calculated by the curves of hydrochloric stachydrine and hydrochloric leonurine, respectively. Results Good linear relationship between the masses and the peak areas of hydrochloric stachydrine and hydrochloric leonufine in the ranges of 0.18 - 1.08 rag( r = 0. 99998 ) and 7 - 41 μg( r = 0. 99998 ), respectively. The average recovery were 99.6% and 99.8% , respectively and RSD were both less than 2.0%. Conclusion The method is simple, rapid, accurate, precise and reliable for the determi- nation of hydrochloric stachydrine and hydrochloric leonurine in motherwort granules.
