TCF7L2基因rs11196218多态性与中国汉族人群T2DM关联的Meta分析

摘　　要：目的:综合评价转录因子7样类似物2(Transcription factor 7 like 2)基因rs11196218多态性与中国汉族人群2型糖尿病(type 2 Diabetes mellitus,T2DM)的关联。方法:通过文献检索收集2015年3月20日以前完成或发表的中国人TCF7L2基因rs11196218多态性与T2DM相关性的病例对照研究或队列研究文献,剔除不符合要求的文献,以漏斗图分析入选文献的发表偏倚,Meta分析采用Revman 5.3版软件,对各入选研究的结果进行一致性检验和数据合并,计算总OR值和95%CI;对各研究结果进行不同分析模型和样本含量的敏感性分析,评价Meta分析结果的稳定性。结果:检索筛选到相关文献11篇纳入研究,得到7 577个T2DM病例和13 072个对照者;异质性检验显示各研究间存在统计学意义的异质性(Q=19.47,I2=49.0%,P=0.03);TCF7L2基因rs11196218风险等位基因(G/A)的分布频率在T2DM组和对照组之间的差异无统计学意义(P=0.17),其合并OR值为1.05(95%CI:0.98,1.14),提示TCF7L2基因rs11196218多态性G/A等位基因与中国汉族人群T2DM不存在关联;发表偏倚分析和敏感性分析结果证实上述Meta分析结果稳定、可靠。结论:TCF7L2基因rs11196218多态性可能不是中国汉族人群T2DM的遗传位点。Objective： To evaluate the relationship of Transcription factor 7-like 2 rs11196218 polymorphisms and type 2Diabetes mellitus（ T2DM） in Chinese han population. Methods： Collection literature of case-control study or cohort study of TCF7L2 rs11196218 polymorphism and correlation of T2 DM in China which are finished and published before March20,2015. Excluding un-requirement literatures,testing publish bias of requirement literatures with funnel. Using Revman5. 3 software in meta analysis,testing the consistency and amalgamation date,calculating OR and 95% CI; Every research results were sensitive analyzed in different analysis model and sample content,evaluation the stability of Meta analysis. Results： 9 literatures had been collected,include 7 577T2 DM cases and 13 072 collator; heterogeneity test show that heterogeneity is exists between every researches（ Q = 19. 47,I2= 49. 0%,P = 0. 03）; frequency distribution of risk allele of TCF7L2 rs11196218（ G / A） have no statistical significance in T2 DM cases and collator（ P = 0. 17）,OR is 1. 05（ 95% CI： 0. 98,1. 14）,It is suggested that the polymorphism of rs11196218 site in TCF7L2 gene and T2 DM in Chinese han population may no correlation. Publish bias analysis and sensitive analysis show that Meta analysis result is stable and reliable. Conclusion： TCF7L2 rs11196218 polymorphisms may not be the T2 DM genetic loci in Chinese population.
