摘 要:以壳聚糖、咪唑-1-乙酸为原材料,1-乙基- (3-二甲基氨基丙基)碳酰二亚胺盐为引发剂、N-羟基丁二酰亚胺为偶联剂,在2-(N-吗啡啉)乙磺酸(MES)缓冲体系中将咪唑-1-乙酸接枝到壳聚糖中,冷冻干燥得到具有pH 值响应性的咪唑-1-乙酸聚糖(IACS).用傅里叶变换红外光谱(FT-IR)、核磁共振(1 H-NMR)、元素分析(EA)、场发射扫描电镜(FESEM)等方法对产物进行表征.结果表明,咪唑-1-乙酸成功接枝到壳聚糖的氨基上;当壳聚糖分子量相同时,随着咪唑-1-乙酸/壳聚糖摩尔比的增加,取代度升高,取代度落在5.52%-7.76%,IACS支架孔径尺寸分布在50-150μm 之间,呈现三维状结构. 当咪唑-1-乙酸壳聚糖溶液pH 值从4.0增加大6.5时,体系的Zeta电位从+64.2mV 降低到+24.2mV.综上所述,咪唑-1-乙酸壳聚糖支架有望为生物矿化和骨组织生长提供良好的模板.Chitosan used as target polymer and was modified with imidazole-1-acetic acid. 1-(3-dimethylamino propyl)-3-ethyl carbodiimide as initiator and N-hydroxysuccinimide as the coupler,the imidazole-1-acetic acid was grafted to chitosan. A series of imidazole-1-acetic acid modified chitosan( IACS) with different substitution degrees were prepared by using the method of freeze-casting and controlling the amount of Imidazole-1-acetic acid and other conditions. Chemical structure and morphology of IACS were characterized through Fourier transform infrared spectrometer( FT-IR),1H-NMR spectroscopy,elemental analysis( EA),field emission scanning electron microscopy( FESEM) and other methods. The results showed that imidazole-1-acetic acid successfully grafted onto the amino groups of chitosan,with increasing of the imidazole-1-acetic acid dosage,the degree of substitution increased(5. 52%-7. 76%),the pore size arranging in 50-150μm,presenting three-dimensional structure. The Zeta value of IACS decreased from + 64. 2 mV to + 24. 2 mV with increase of p H value,which proved that IACS has p H responsibility. Based on the above results,it indicted that IACS could provide a good template for biomineralization and be applied in cartilage tissue engineering.
