摘 要:目的探讨甲苯暴露对小鼠肝脏、肺脏、肾脏、心脏、脑组织等靶点氧化损伤情况,探寻甲苯对机体作用的敏感、早期检测指标。方法将18只染毒小鼠根据体重随机分成甲苯高剂量组、低剂量组和对照组,采用静式吸入染毒方式,每天染毒2 h,共染毒30 d。实验结束后,测定小鼠各脏器中的氧化损伤指标。结果各脏器中SOD、GSH含量随染毒浓度的增加而减少(P<0.05),MDA含量随染毒浓度的增加而增加(P<0.05),染毒组和对照组的肺脏脏器系数明显不同(P<0.05),以上差异均有统计学意义。甲苯高剂量染毒组肝脏、心脏和肺脏中GSH、SOD含量均低于对照组(P<0.05),脑组织中GSH含量低于对照组(P<0.05),肾脏中GSH含量低于对照组(P<0.05);低剂量组心组织中SOD含量低于对照组(P<0.05);以上差异均具有统计学意义。结论不同浓度甲苯暴露下,小鼠各脏器均有氧化损伤;随着甲苯浓度的增加:小鼠肝脏、心脏、肺脏的GSH、SOD含量显著降低,MDA含量显著升高,且肝脏最为敏感;小鼠肾脏、脑组织的GSH含量逐渐降低,MDA含量逐渐升高。Objective To observe the effect of oxidative damage of toluene exposure on liver, lungs, kidney, heart, brain and other target in mice, and find the sensitive target organs of the body after long - term exposure to low concentration of toluene in order to explore the impact of toluene and early detection target. Methods Eighteen mice were randomly divided into three groups according to the weight as high dose group, low dose group and control group. The treatments were con- ducted by static inhaling toluene, two hours per day for thirty days, and determination of the organs of mice induced by oxi- dative damage indicator. Results The SOD and GSH content in various organs decreased with increasing exposure dose ( P 〈 0. 05 ), MDA content increased with increasing exposure dose (P 〈 0. 05), a significant difference of lung organ coef- ficient between the exposure group and control group(P 〈 0. 05). The SOD, GSH contents of the liver,the lungs and heart in the high dose group were less than those in the control group( P 〈 0. 05 ), and GSH contents of the brain and kidney were also less than those in the control group ( P 〈 0. 05 ) ; The SOD content of the heart in the low dose group was less than that in the control group( P 〈 0.05 ). Conclusion Each organ has the oxidative damage in mice with different concentrations of toluene exposure ; The contents of the GSH and SOD of the liver, the heart and lungs in mice decrease significantly as the increasing of the concentration of toluene, and MDA increases significantly, the liver is the most sensitive; The content of the GSH of kidney and brain in mice gradually reduces, and the MDA increases gradually.
