摘 要:以新疆大叶紫花苜蓿继代培养6~8周的胚性愈伤组织为材料,研究不同辐射剂量的γ射线(60Co)辐射诱变对不同盐浓度和胁迫时间下的胚性愈伤组织芽分化率的影响以定向筛选耐盐变异体.在体细胞胚发生时期的细胞学观察发现,继代培养6~8周的胚性愈伤组织分化出大量的胚性细胞,是进行y射线辐射处理的最佳时期;比较0~100Gy辐射剂量分别在0%~1.0%盐浓度胁迫1~3个月下的胚性愈伤组织芽分化率发现,随着盐处理时间的延长,不同辐射剂量处理的芽分化率均呈现缓慢下降的趋势;回归分析表明,新疆大叶苜蓿体细胞胚的适宜辐射强度为2~8Gy,80Gy剂量的少量突变苗生长良好;辐射诱变具有不定向性,得到了耐盐性高达1.0%的突变株.The embryonic callus of Xinjiang Daye alfalfa subcultured in 6-8 weeks was used as material, the effect of 7 irradiation(60Co)in different radiation dose under salt-tolerance of different concentration on differentiation rate of callus were studied in order to screen salt-tolerance variant directionally. It was found that embryonic callus subcultured in 6 -8 weeks differentiated large number of embryonic cells, which was the appropriate time for 7-radiation treatment. Number of buds under 0-100Gy radiation dose decreased slowly with increasing hours of NaC1 treatment; Regression analysis showed that the suitable dose of radiation for somatic embryo was 2-8Gy; some mutated seedlings under 80Gy radiation dose grow better; mutation breeding has the non directionality;mutants for salt tolerance of 1.0 %So NaC1 were obtained.
