摘  要:目的研究脑脊液诱导的骨髓源性神经样细胞(BMSC—Ns)移植的安全性。方法将40只4。6周龄裸鼠采用随机数字表法分为5组(n=8),其中致瘤性实验3组:阳性对照组(A组),接种Hela细胞(2×107/0.2m1);实验组(B组),将BMSC—Ns按2×107/0.2ml接种至裸鼠肋部皮下;阴性对照组(C组),注射等体积生理盐水。接种后饲养1个月,大体观察肿瘤形成情况,1个月后处死动物取注射处局部皮肤、心脏、肝脏、肠道和肺组织,行H—E染色观察病理学改变。全身毒性实验2组:实验组(D组),裸鼠尾静脉注射大鼠BMSC—Ns(5x106/0.2m1);阴性对照组(E组),注射等体积生理盐水。注射后即刻观察动物临床反应,以后每天观察1次,连续观察15天;将动物处死,采集血液标本行血液学检查。结果BMSC—Ns接种于裸鼠肋部皮下1个月后未见接种部位肿瘤形成,局部皮肤、心脏、肝脏、肠道和肺组织病理学检查未见异常;尾静脉注射BMSC—Ns后动物一般情况无异常,至第15天裸鼠全部成活,血常规结果无异常。结论皮下接种或者尾静脉移植CSF诱导的BMSC—Ns对裸鼠是安全的,无致瘤性和全身毒性。Objective To assess the safety of transplantation of CSF-induced bone marrow-derived neuron-like cells (BMSC-Ns). Methods A total of 40 nude mice (4-6 weeks old) were randomly divided into five groups (n= 8). Among them were three groups which underwent tumorigenicity experiments, namely a positive control group (group A) inoculated with Hela cells (2~ 107/0.2 ml), an experimental group (group B) that were subcutaneously inoculated with CSF-induced BMSC-Ns (2× 107/0.2 ml) into the ribs of nude mice and a negative control group (Group C) that were injected with the same volume of normal saline. After inoculation, the development of tumor was observed for one month. Then the animals were sacrificed and the liver, heart, lung, intestine and the patch of the skin of injection site were removed for pathological analysis by HE staining. There were another two groups involved in systemic toxicity experiments, namely an experimental group (Group D) injected with CSF-induced BMSC-Ns (5×106/0.2 ml) via the tail vein and a negative control group (E group) injected with the same volume of normal saline. Then, the responses of animals were observed immediately after injection. The responses were then recorded once per day for 15 consecutive days. The animals were sacrificed on Day 15 before collection of blood samples for hematological examination. Results No tumor was developed at the injection site of CSF-induced BMSC-Ns one month after inoculation. Meanwhile, pathological examination showed no abnormalities in the local skin, heart, liver, intestine and lung. The general situation and blood routine examination was normal in the nude animal after injection of BMSC-Ns, while all animals were live until scarified. Conclusion Subcutaneous inoculation or transplantation of CSF-induced BMSC-Ns via the tail vein are safe for nude mice, without neither tumorigenity nor systemic toxic effects.
