摘  要:目的:探讨苯丙胺对额叶皮质部位AKT信号通路上游信号分子的影响。方法:选取健康雄性成年SD大鼠50只,分为空白组、生理盐水组和苯丙胺(AMPH)组。用旷场(open field)系统测试大鼠的行为学变化,用Western blot分析AKT信号通路上游信号分子的变化,用免疫组织化学观察相应分子阳性细胞的变化。结果:注射苯丙胺后大鼠活动度增加,且出现刻板行为。AMPH组大鼠额叶皮质pm TOR含量比空白组及生理盐水组减少,且pm TOR阳性细胞的数量也相应减少,而p PP2A和PHLPP没有显著性变化。结论:苯丙胺抑制AKT信号通路可能与减少pm TOR的表达密切相关。Objective : To investigate the changes of signal molecules in the upstream of AKT pathway in the frontal cortex of rats under treatment of amphetamine. Methods: Fifty healthy adult male SD rats were randomly divided into the blank, saline, and amphetamine (AMPH) groups. Rat behaviors were tested by open filed system. The Western blot was used to analyze the changes of upstream molecules of AKT pathway in the frontal cortex, and the positive cell changes of the pathway were observed using immunohistochemistry. Resu/._ts_: Amphetamine increased the rat activities and resulted in stereotypic behaviors. The expression of phosphorylated mammalian target of rapamycin (pmTOR) in the frontal cortex in AMPH group was decreased,as compared with that in the blank and saline groups. The number of pmTOR postivie cells in the frontal cortex was also significantly decreased in the AMPH group. However,there was no significant difference between groups in the expression of pPP2A and PHLPP. Conclusion : Decreased expression of pmTOR may contribute to amphetamine - induced inhibition of AKT pathway.
