摘　　要：目的：分析LIN28在男性原始生殖细胞（PGCs）中的表达变化。方法：收集男性胚胎/胎儿,胎龄为7周、8周、9周、10周、11-12周、13-14周各3例。分离早期胚胎性腺组织,运用免疫荧光技术分析PGCs中LIN28和OCT4的表达。结果：6组睾丸组织中LIN28和OTC4阳性细胞百分数比较,差异均有统计学意义（F=15.651,6.501,P均〈0.05）。胎龄第7-14周,OCT4一直定位于PGCs的细胞核中;胎龄第8-9周时,OCT4在PGCs中的表达低于第7周,第10-12周OCT4表达再次升高,而在第13周后,OCT4表达逐渐降低。从胎龄第7周开始直至第14周,PGCs中LIN28蛋白的表达逐渐增强,在胎龄第13-14周达到最高峰。在定位上,在胎龄第7周时LIN28表达于PGCs胞核内;在胎龄第8-9周时,PGCs胞核和胞质中均可观察到LIN28的表达;而在胎龄第10周以后,部分PGCs仅胞质中表达LIN28;在胎龄第11-14周时LIN28均定位于PGCs的胞质中。结论：LIN28可能是一个可行的、比较稳定的雄性生殖细胞标记因子。Aim： To explore the expression of LIN28 in the male primordial germ cells（ PGCs）. Methods： The male embryo / fetus were collected（ gestational age of 7 weeks,8 weeks,9 weeks,10 weeks,11- 12 weeks,13- 14 weeks was3 cases,respectively） to obtain human fetal testicular tissue. Immunofluorescence technique was used to detect the expressions of LIN28 and OCT4 in PGCs. Results： There were significant differences in the expressions of LIN28 and OCT4 among the 6 groups（ F = 15. 651,6. 501, P〈0. 05）. At the gestational age of 7- 14 weeks,OCT4 was located in the nucleus of PGCs. The expression of OCT4 in PGCs at the gestational age of 8- 9 weeks was lower than that at the gestational age of7 weeks,and it increased at the gestational age of 10- 12 weeks,decreased after the gestational age of 13 weeks. From the gestational age of 7 to 14 weeks,the expression of LIN28 in PGCs increased gradually,and reached to the peak at the gestational age of 13- 14 weeks. At the gestational age of 7 weeks,LIN28 expressed within the nuclei of PGCs. At the gestational age of 8- 9 weeks,LIN28 expressed within the nucleus and cytoplasm of PGCs,LIN28 only expressed in the cytoplasm in partial PGCs at gestational age of 10 weeks. At the gestational age of 11- 14 weeks,LIN28 was positioned in the cytoplasm of PGCs. Conclusion： LIN28 may be a feasible and relatively stable male primordial germ cell marker.
