摘  要:目的:探讨葡萄籽原花青素(Grape Seed Proanthocyanidin Extract,GSPE)的抗肝癌活性及可能的机制。方法:以离体人肝癌Hep G2细胞及在体荷瘤小鼠为研究对象,利用MTT法及流式细胞术评价葡萄籽原花青素体外对人肝癌Hep G2细胞增殖及凋亡的影响。建立人肝癌Hep G2细胞裸鼠皮下种植瘤模型,腹腔注射GSPE,观察其对体内种植瘤模型的作用。结果:GSPE对人肝癌Hep G2细胞具有明显的增殖抑制作用,并将人肝癌Hep G2细胞阻滞在S期。在体内则可显著抑制皮下移植瘤的生长。进一步研究结果显示,GSPE可降低Hep G2细胞中活性氧(Reactive Oxygen Species,ROS)含量,同时可明显增高Hep G2细胞中超氧化物歧化酶(Superoxide Dismutase,SOD)的活性。结论:GSPE可明显抑制体内外肝癌的生长及发展,这可能与其较强的抗氧化活性有关。Objective: To explore the anti-hepatocarcinoma activity and radical mechanism of grape seed Proanthocyanidin extract. Methods:To evalue the effects of grape seed Procyanidins on proliferation and apoptosis of human hepatoma cell line HepG2 cells using MTY method and flow cytometric in human hepatoma HepG2 cells in vitro and in vivo with tumor-bearing mice. The nude mices were injected subcutaneously implantation model to establish human hepatoma cell line HepG2 cells,which were intraperitoneally injected GSPE and observed its effect on in vivo tumor models. Results: GSPE had obvious inhibition on the proliferation of human hepatoma HepG2 cells, and blocked human hepatoma cell line HepG2 cells in S phase,which can significantly inhibited the growth of subcutaneous tumors in vivo. GSPE could reduce the reactive oxygen species in HepG2 cells ,and increased the superoxide dismutase activity in HepG2 cell. Conclusion: GSPE can obviously inhibit growth and development in vitro and in vivo liver cancer, which may is associated with its strong antioxidant activity.
