摘 要:目的探究ST2基因单核苷酸多态性(SNPs)与糖尿病发病风险的相关性。方法选取2008~2010年收治的1 726例资料完整的住院患者,将其分为糖尿病组(409例)和非糖尿病组(1 317例),利用HapMap数据库选取10个ST2基因标签SNPs,采用TagMan荧光探针法检测位点多态性,应用Logistic回归模型分析SNPs与糖尿病发病风险之间的相关性。结果 1所选10个ST2基因SNPs中rs10515922基因型频数在糖尿病组和非糖尿病组的分布差异具有统计学意义(P=0.035);二元Logistic回归分析在校正年龄、性别、体重指数(BMI)、腰围、甘油三酯、总胆固醇、吸烟、饮酒、高血压病及冠心病后,相比其TT基因型,TC、TC+CC基因型糖尿病风险分别下降34%和33%(95%CI 0.493~0.883,P=0.005;95%CI0.506~0.889,P=0.005);不同rs10515922基因型患者之间的BMI、腰围、甘油三酯、总胆固醇水平差异无统计学意义。2与非糖尿病组相比,糖尿病组更易患冠心病和高血压病(62.6%vs.45.3%,P〈0.01;57.2%vs.37.5%,P〈0.01)。结论 ST2基因rs10515922可能是汉族人群糖尿病的独立保护因素。Objective To investigate the relationship between ST2 gene polymorphisms and diabetes mellitus(DM)risk in the Han population.Methods A total of 1 726inpatients(Han Chinese)with complete medical records who were treated in our hospital between 2008 and 2010were enrolled in the study.They were divided into DM group(n=409)and non-DM group(n=1 317).Ten tagger single nucleotide polymorphisms(SNPs)in ST2 gene region were selected by using HapMap database.The alleles were determined by using the TaqMan fluorescent probes.Logistic regression model was used to analyze the association between the 10 SNPs and DM.Results 1The distribution of rs10515922 genotype frequencies was significantly different between the two groups(P=0.035);binary logistic analysis showed that after adjusting for age,gender,body mass index(BMI),waistline,triglyceride and total cholesterol levels,smoking,drinking,hypertension and coronary heart disease,the risk for DM was decreased by 34% and 33%,respectively,in patients with TC and TC+CC genotypes when compared with those with TT genotype[95% CI0.493-0.883,P=0.005;95% CI 0.506-0.889,P=0.005);no significant difference was noted in BMI,waistline,triglyceride and total cholesterol levels between patients with rs10515922 genotypes.2Patients in DM group compared to non-DM group had higher prevalence of hypertension and coronary heart disease(57.2%vs.37.5%,P〈0.01;62.6%vs.45.3%,P〈0.01).Conclusion SNP rs10515922 in ST2gene region is probably an independent protective factor for DM in the Han Chinese population.
