摘 要:目的:研究草酸钙肾结石大鼠肾组织TH蛋白的表达,并用中药温肾化石汤调节,探讨该方防治肾结石复发的作用机制。方法:雄性SD大鼠60只,随机分为对照组、肾结石组、温肾化石汤组,每组各20只,肾结石组和温肾化石汤组以含有0.5%的乙二醇和0.5%的氯化铵的混合溶液致石剂代替饮用水,温肾化石汤组同时每日灌服中药温肾化石汤,二周后处死大鼠,解剖观察肾脏剖面结石形成,并用免疫组织化学法测定肾组织TH蛋白的相对表达量。结果:肾结石组肾脏成石程度最重,与温肾化石汤组比差异显著,对照组肾组织TH蛋白表达最强,其次为温肾化石汤组,肾结石组,三组相比差异显著。结论:TH蛋白可以抑制肾结石形成,中药温肾化石汤可以恢复大鼠肾组织TH蛋白表达量,推测此可能为该方防治肾结石复发的重要作用机制。To study the expression of TH-protein in Renal Tissue of Rats with Calcium Oxalate Stones and the regulation of Wenshen Huashi Decoction, and explore the mechanism of prevention and cure the recurrence of kidney stones for this therapy.Method:60 male SD rats were randomly divided into three groups, which were control group, kidney stone group, Wenshen Huashi Decoction group, each group of 20 rats.Instead of drinking water, kidney stones group and Wenshen Hua shi Decoction group drunk the mixed solution which lead to stone water that were contain 0.5%of glycol and 0.5%of ammonium chloride, at the same time Wen Shen Hua Shi De-coction group fed the Wenshen Huashi Decoction daily.Rats were killed after two weeks, observed the formation of stone on the kidney profile, and then tested the relative expression amount of TH-protein in renal tissue which used the immunohistochemistry method. Results:Kidney stone group were the most severity in stone formation, and there were a significant difference than the Wenshen Huashi Decoction group.TH-protein were strongest expressed in contral group,and then Wen shen Huashi Decoction group and kidney stone group, significant difference in TH-protein expression among three groups.Conclusion:TH-protein can inhibit the formation of kid-ney stones, Wenshen Huashi Decoction can restore the TH-protein expression in rats kidney tissue, thus speculated that this may be the important role in the mechanism of prevention and cure the recurrence of kidney stones for Wenshen Huashi Decoction.
