摘 要:目的探讨细胞间黏附分子-1(ICAM-1)在油酸所致家兔急性肺损伤(ALI)中的作用。方法健康新西兰家兔30只随机分为空白组、对照组和实验组,每组10只。实验组家兔通过耳缘静脉注射油酸复制油酸性ALI模型,对照组家兔经耳缘静脉注射等量生理盐水,空白组家兔不进行任何耳缘静脉注射。记录3组家兔的呼吸曲线,酶联免疫吸附测定法测定外周血和支气管肺泡灌洗液(BALF)中ICAM-1水平,免疫组织化学法测定家兔肺组织中ICAM-1的表达。结果实验组家兔吸气时间、呼气时间显著短于对照组和空白组(P<0.05),呼吸幅度显著低于对照组和空白组(P<0.05),呼吸频率显著高于对照组和空白组(P<0.05)。实验组家兔动脉血氧分压(Pa O2)显著低于对照组和空白组(P<0.01),实验组家兔动脉血二氧化碳分压(Pa CO2)显著高于对照组和空白组(P<0.01)。对照组与空白组家兔Pa O2、Pa CO2比较差异均无统计学意义(P>0.05)。实验组家兔外周血和BALF中ICAM-1水平显著高于对照组和空白组(P<0.01)。对照组与空白组家兔外周血和BALF中ICAM-1水平比较差异均无统计学意义(P>0.05)。实验组家兔肺组织中ICAM-1蛋白的表达显著高于对照组和空白组(P<0.05)。结论 ICAM-1可能在油酸性ALI的发生发展中起重要作用。Objective To explore the role of intercellular cell adhesion molecule-1( ICAM-1) in acute lung injury(ALI) induced by oleic acid in rabbits. Methods Thirty New Zea Land rabbits were randomly divided into blank group,control group and experiment group,ten rabbits in each group. The rabbits in experiment group were injected with oleinic acid by ear vein to make ALI models. The rabbits in control group were injected with equivalent normal saline by ear vein. The rabbits in blank group were not injected with anything. The respiratory curves of rabbits were recorded in the three groups. The level of ICAM-1 in peripheral blood and bronchoalveolar lavage fluid( BALF) was detected by enzyme-linked immunosorbent assay.The expression of ICAM-1 in lung tissue of rabbits was detected by immunohistochemical method. Results Compared with control group and blank group,the inspiratory time and expiration time was significantly shorter( P〈0. 05),and the respiratory amplitude was significantly lower( P〈0. 05),but the respiratory frequency was significantly higher in experiment group( P〈0. 05). The Pa O2 in experiment group was significantly lower than that in control group and blank group( P〈0. 05). The Pa CO2 in experiment group was significantly higher than that in control group and blank group( P〈0. 05). There was no significant difference in Pa O2 and Pa CO2 between control group and blank group( P〉0. 05). The ICAM-1 level in peripheral blood and BALF in experiment group was significantly higher than that in control group and blank group( P〈0. 05). There was no significant difference in the level of ICAM-1 in peripheral blood and BALF between control group and blank group( P〈0. 05). The expression of ICAM-1 protein in lung tissue of rabbits in experiment group was significantly higher than that in control group and blank group( P〈0. 05). Conclusion ICAM-1 may play an important role in the development of ALI induced by oleic acid.
