摘 要:目的研究盐酸小檗碱与吴茱萸碱联用抑制乳腺癌转移作用及机制。方法将人类乳腺癌(MDA-MB-231)接种于24孔板,随机分为4组,对照组、盐酸小檗碱组、吴茱萸碱组及吴茱萸碱小檗碱联用组;用酶联免疫吸附试验(ELISA)测定MDA-MB-231细胞白细胞介素-8(IL-8)表达;用细胞划痕实验测定MDA-MB-231细胞的迁移能力。结果不同浓度盐酸小檗碱均能抑制不同浓度吴茱萸碱诱导的MDA-MB-231细胞IL-8表达的增加。盐酸小檗碱和吴茱萸碱均能显著抑制MDA-MB-231细胞迁移,盐酸小檗碱与吴茱萸碱联用能够协同抑制MDA-MB-231细胞的迁移。结论盐酸小檗碱与吴茱萸碱联用能够抑制吴茱萸碱诱导乳腺癌转移的潜在不良反应。Objective To investigate the effect and underlying mecha-nism of combination of berberine hydrochloride and evodiamine on breast cancer metastasis. Method MDA -MB -231 cells were seeded in 24-well plates and randomly divided into the following groups, namely control group, berberine-treated group, evodiamine-treated group and the combination of berberine hydrochloride and evodiamine group.Inter-leukin-8 ( IL-8 ) contents were measured by enzyme-linked immu-nosorbent assay( ELISA) method.The migration ability was measured by wound healing assay.Results The up-regulation of IL-8 expression induced by evodiamine could all be inhibited by different concentrations of berberine hydrochloride in MDA -MB -231 cells.Combination of berberine hydrochloride and evodiamine could trigger synergistic inhibi-tion of MDA-MB-231 cell migration.Conclusion Berberine hydro-chloride could counteract the potential adverse reaction of evodiamine on cancer metastasis of MDA-MB-231 cells.
