摘　　要：病毒分离是流感病原监测的金标准。实际工作中,特别是刚开展工作的实验室会遇到许多问题和困惑。文章就病毒培养中样本质量、接种细胞、胎牛血清、收获时间和鉴定实验中稀释液、血凝效价测定、4个单位抗原配置、反应温度等一些主要影响因素及注意事项进行了归纳和总结。Virus isolation is the gold standard test for flu surveillance. However, there would be many problems in our practical work, particularly to those labs whose work has just been carried out. In this paper, a serious of main influence factors and precautions in both viral cultivation （such as sample quality, ceils for isolation, the fetal bovine serum and harvest time） and virus identification （including buffer solution, determination of titer of hemagglutination, four units of antigens preparation and reaction temperature） were summarized.
