摘  要:目的:探讨微管相关蛋白(Doublecortin)在幼犬脑组织不同部位的表达,进一步研究出生后家犬神经发生和神经元迁移机制。方法:4只出生后幼犬给以正常饮食饲养2周,4%多聚甲醛溶液灌注取脑,通过免疫组织化学方法检测微管蛋白在幼犬脑皮层、室管膜下区、白质和海马的分布表达情况,观察不同部位新生神经元的形态和突起的伸展走行分布。结果:微管蛋白在脑皮层、室管膜下区、白质和海马均有分布,室管膜下区和海马齿状回表达最高,神经元突起表达明显,伸展方向较为一致,朝向大脑皮层。脑皮层第Ⅱ层以下层面均有表达。结论:家犬出生后早期脑组织仍有新生神经元产生,神经元生发区位于室管膜下区和海马齿状回,室管膜下区和齿状回新生神经元在迁移的过程中分化成熟,最终迁移至脑皮层,完成神经系统发育。Objective:To study the expression of Doublecortin in the brain of young canis familiaris,and further explore the neurogenesis and neuron migration mechanisms of canis familiaris at early stage.Methods:4postnatal young canis familiaris were fed with normal diet for2 weeks.The brains were extracted by 4% paraformaldehyde solution perfusion.The expression and distribution of Doublecortin in cerebral cortex,subependymal zone,brain white matter and dentate gyrus were detected by immunohistochemistry;the morphology and distribution of newly-born neurons and neurite were observed in different brain regions.Results:Doublecortin are expressed in cerebral cortex,subependymal zone,brain white matter and dentate gyrus;the highest expression level appeared in subependymal zone and dentate gyrus.Neurite were distinctively stained,and stretched consistently to the cerebral cortex.Doublecortin were expressed extensively in layerⅡ and below the layerⅡ of the cerebral cortex.Conclusion:At the postnatal early stage,abundant newly-born neurons still occurred in the brain regions of the young canis familiaris,The same as other mammals,Neurogenesis area is located in subependymal zone and dentate gyrus.Newly-born neurons in the two regions are gradually differentiated to maturation in the course of migration,and are located in the cerebral cortex eventually,complete development of the central nervous system(CNS).
