摘  要:目的探讨益心泰颗粒对慢性心力衰竭(chronic heart failure,CHF)兔Ca2+-CaN-NFAT3信号通路的影响。方法用阿霉素建立CHF兔模型,将造模成功兔分为模型组、益心泰低、中、高剂量组和氯沙坦钾组;另设正常对照组。灌胃4周后观察心肌组织病理结构,并检测心肌细胞内钙离子浓度,心肌组织钙调神经磷酸酶(CaN)、T细胞活化因子3(NFAT3m RNA)及其蛋白和磷酸化水平。结果与正常对照组比较,模型组心肌细胞内[Ca2+]i含量、心肌组织CaN水平、NFAT3、p-NFAT3及NFAT3m RNA表达显著升高(P﹤0.01)。与模型组比较,益心泰低剂量组心肌组织NFAT3、NFAT3m RNA表达降低(P﹤0.05),心肌细胞内[Ca2+]i含量、心肌组织CaN水平及p-NFAT3表达显著降低(P﹤0.01),而益心泰中、高剂量组和氯沙坦钾组心肌细胞内[Ca2+]i含量、心肌组织CaN水平、NFAT3、p-NFAT3及NFAT3m RNA表达显著降低(P﹤0.01)。结论益心泰颗粒抑制慢性心力衰竭时Ca2+-CaN-NFAT3信号通路激活,可能是其治疗慢性心衰的主要机制之一。Objective To explore the effects of Yixintai granule on the Ca2+-CaN-NFAT3 signal pathway in rabbits with chronic heart failure(CHF). Methods The CHF rabbit models were established with Adriamycin. Then the rabbit models were divided into model group, high-dose, middle-dose, low-dose of Yixintai groups, and Losartan Potassium group. The separate normal group was designed. After 4 weeks of gavage administration, the changes of myocardial tissue pathological structure were observed, and the concentration of myocardial intracellular calciumion, the levels of CaN, NFAT3 m RNA, and its protein and phosphorylation in myocardial tissue were measured. Results Compared with the normal group, the concentration of myocardial intracellular calciumion, the levels of CaN, NFAT3, p-NFAT3, and NFAT3 m RNA were increased significantly(P ﹤ 0.01). Compared with the model group, the expressions of NFAT3 and NFAT3m RNA in myocardial tissue were decreased(P﹤ 0.05), the concentration of myocardial intracellular calciumion, the activity of CaN and the expression of p-NFAT3 in myocardial tissue of the low-dose Yixintai group were decreased significantly(P ﹤0.01); but the concentration of myocardial intracellular calciumion, the levels of CaN, NFAT3, p-NFAT3, NFAT3 m RNA in myocardial tissue of the middle-dose, high-dose of Yixintai group and Losartan Potassium group were decreased significantly(P﹤0.01). Conclusion The mechanism of Yixintai improving the rabbits with chronic heart failure granule maybe pertinent to inhibiting the Ca2+-CaN-NFAT3 signal pathway activation.
