摘 要:目的观察大鼠哮喘血清诱导气道上皮细胞表达T细胞成熟相关蛋白(T-lymphocyte maturation associated protein T,MAL)以及用柴朴汤血清干预对气道上皮细胞MAL表达的影响。方法健康SD雄性大鼠64只,随机抽取32只分为柴朴汤高(3.00 g/kg)、中(1.50 g/kg)、低(0.75 g/kg)剂量组、生理盐水组,每组8只,连续给药7 d,每天2次,药后经心脏采血分离血清分别得到柴朴汤高、中、低剂量血清组、正常血清组。剩余32只以卵蛋白、氢氧化铝造哮喘模型,通过心脏采血后分离得哮喘血清,并取肺组织进行HP染色。取正常大鼠气道上皮细胞培养,分别设正常血清组、柴朴汤高剂量血清+哮喘血清组(下称柴哮高组)、柴朴汤中剂量血清+哮喘血清组(下称柴哮中组)、柴朴汤低剂量血清+哮喘血清(下称柴哮低组)、正常血清+哮喘血清组(下称盐哮血清)及哮喘血清组,诱导各组上皮细胞,用半定量逆转录-聚合酶链反应(Reverse Transcription Polymerase,RT-PCR)检测大鼠气道上皮细胞中MAL的表达,用免疫组织化学(Immunohistochemistry,IHC)检测MAL蛋白在气道上皮细胞中的表达。结果哮喘血清干预后,与正常血清组比较,MAL m RNA和蛋白表达显著降低(P<0.05),盐哮血清组较正常血清组也明显降低(P<0.05);柴哮高、中、低组较盐哮血清组及哮喘血清组有所增强(P<0.05),柴朴汤血清作用后,气道上皮细胞中MAL的表达明显上调。结论哮喘血清可诱导气道上皮细胞中MAL的表达下调,柴朴汤上调MAL的表达,维护气道上皮细胞的正常形态和功能。Objective To observe the expression of T-lymphocyte maturation associated protein T(MAL) in airway epithelial cells induced by asthma serum and the Chaipu decoction's intervention on the expression of MAL in the airway epithelial cell. Methods 32 rats were randomly picked out from 64 healthy male SD rats, and assigned into Chaipu decoction high dose(3.00 g/kg), mid dose(1.50 g/kg), low dose(0.75 g/kg)and normal saline by intragastric administration for 7 days, twice a day. After treatment, serum was separated from blood which was from rat heart, then the high dose,mid dose,low dose Chaipu decoction serum groups and normal serum group were acquired respectively. The other 32 rats were established the asthmatic rat models induced by ovalbumin(OVA) and aluminum hydroxide. After treatment,the asthma serum was collected from the heart, and the lung tissue was obtained and stained by HE. The normal airway epithelial cells were cultured and divided into normal serum group, Chaipu decoction high dose and asthma serum group(HCDAS group), medium Chaipu dose and asthma serum group(MCDAS group), low Chaipu dose group and asthma serum group(LCDAS group), normal serum and asthma serum group(NAS group), asthma serum group. The expressions of MAL m RNA and protein in airway epithelial cells were detected by the RT-PCR and immunohistochemical method. Results Under the intervention of asthma serum, the MAL m RNA and protein expression levels in asthma serum group were lower than normal serum group(P〈0.05), the expressions in NAS group were decreased obviously(P〈0.05). The HCDAS MCDAS and LCDAS groups were significantly higher than those in NAS group and asthma serum group(P〈0.05). The expression of MAL was improved after Chaipu decoction serum intervention. Conclusion The asthma serum could induce the down-regulated expression of MAL in airway epithelial cells.
