摘  要:研究建立了吡虫啉与噻虫嗪的间接竞争酶联免疫吸附分析(ELISA)方法,并用于粮食样品中农药残留检测,采用杂交瘤技术制备了吡虫啉和噻虫嗪的单克隆抗体,用方阵法筛选包被抗原与抗体的浓度,在优化条件下建立了两种杀虫剂的间接竞争抑制曲线,吡虫啉间接竞争ELISA方法的抑制中浓度IC50=6.5ng/mL,最低检测限IC10=0.68ng/mL,噻虫嗪间接竞争ELISA方法的抑制中浓度IC50=0.57ng/mL,最低检测限IC10=0.08ng/mL;粮食样品中吡虫啉和噻虫嗪的添加回收率在87%~112%之间。对本试验所采集的12个粮食样品进行检测,均未检测到农药残留。在实际生产、生活中,粮食中的有机磷农药污染已经对人们的身体健康构成了严重的威胁,有必要采取有效检测有机磷农药的手段,加强对粮食中农药残留检测的力度,能控制、预防人员中毒,对保障人们的安全有重要意义。The indirect competitive enzyme of imidacloprid and thiamethoxam linked immunosorbentadsorption analysis method had been established (ELISA), used for the detection of pesticide residues in grain samples. The monoclonal antibody of imidacloprid and thiamethoxam were prepared by hybridoma technique, screening concentration of coated antigen and antibody with the matrix method, under the optimized condition indirect competitive inhibition curves of two kinds of insecticides, inhibit the indirect competitive ELISA method of imidacloprid concentration in IC50= 6.5 ng/mL, the detection limit of IC10=0.68 ng/mL, inhibition of indirect competitive ELISA method of thiamethoxam in concentration of IC50= 0.57 ng/mL, the detection limit of IC10=0.08ng/mL, imidacloprid and thiamethoxam in grain sample adding recovery rate was 87-112 %. 12 grain samples collected hadn't detected pesticide residues. In actual production and life, pollution of organic phosphorus pesticides in grain has serious thread on the health of people, need to take effective detection of organic phosphorus pesticide means, to strengthen the detection of pesticide residues in grain, to control and prevent the pollution which is very important to guarantee people's safety.
