摘  要:目的:分析阿托伐他汀对自发性高血压大鼠心肌细胞PPAR-γ及GLUT-4表达水平的影响,为临床运用提供理论指导。方法:选择20只自发性高血压大鼠,随机分为A组(10只)与B组(10只),同时选择10只健康大鼠作为对照组成C组。给予A组大鼠阿托伐他汀按照50mg·kg-1·d-1的水准灌胃喂养,同时给予B组、C组大鼠以同量的蒸馏水灌胃。每隔1周测量三组大鼠的收缩压与舒张压值,测量5次,直到第10周;同时检测三组大鼠10周平均的血脂水平与左心室质量指数水平,比较三组大鼠血脂水平差异性;采用免疫组化法及PT-PCR法检测三组大鼠的PPAR-γ及GLUT-4表达水平。结果:A组与B组大鼠的舒张压与收缩压水平较C组大鼠均较高,具有明显差异性(P<0.05),A组与B组大鼠的血压值水平比较,差异性不明显(P>0.05);A组与B组大鼠的TC、TG、HDLC以及LDL-C等血脂水平较C组大鼠均较低,具有明显差异性(P<0.05),A组与B组比较血脂水平无差异性(P>0.05)。三组大鼠的LVM1水平两两比较均具有显著性差异(P<0.05),但B组大鼠的LVM1水平最高,其次是A组,C组大鼠水平最低;干预前,A组与B组大鼠PPAR-γ及GLUT-4表达水平均较C组较低,具有统计学意义(P<0.05),但A组与B组比较差异性不大(P>0.05);干预后,A组大鼠PPAR-γ及GLUT-4表达水平显著提高,与C组大鼠比较差异性不大,但与B组大鼠比较,具有较大差异性(P<0.05)。结论:阿托伐他汀喂养自发性高血压大鼠,能够降低左心室质量指数水平,但对血压、血脂水平无明显影响,这可能与上调PPAR-γ及GLUT-4表达水平有关,具体机制需要进一步研究。Objective: To analyse affect of spontaneously hypertensive rat cardiomyocyte expression of PPAR-γ and GLUT- 4 with atorvastatin,in order to provide theoretical guidance for clinical application.Method: 20 spontaneously hypertensive rats were randomly divided into group A( 10) and group B( 10),10 healthy rats were selected as group C. Group A were feeded with atorvastatin in dose of 50mg·kg-1·d-1by gavaging,group B and group C were feeded with the same amount of distilled water by gavaging. Systolic and diastolic blood pressure values of the three groups were measured every other week until 10 weeks; Lipid levels and left ventricular mass index of three groups were measured for 10 weeks,Lipid levels of three groups were compared; PPAR-γ and GLUT-4 expression levels of three groups were detected by immunohistochemistry and PT-PCR method. Result: Diastolic and systolic blood pressure levels in group A and group B rats were higher than group C rats( P〈0.05),group A and group B median level of blood pressure had no significant( P〉0.05); lipid levels of TC,TG,HDL-C and LDL-C in group A and group B were lower than group C rats( P〈0.05),lipid levels had no relatively difference between group A and group B( P〉0.05).LVM1 levels among three groups were significant differences( P〈0. 05),and group B was the highest,group A was Secondly higher,group C was the lowest; before the intervention,PPAR-γ and GLUT- 4 expression of group A and group B was lower than group C( P〈0.05),but group A and group B were not significant( P〉0. 05); after the intervention,PPAR- γ and GLUT- 4 expression levels was significantly increased,there was not significant compared with group C,but there was significant different compared with group B( P〈0.05). Conclusion: Atorvastatin fed spontaneously hypertensive rats,can reduce the level of left ventricular mass index,blood pressure,but no significant effect on blood lipid levels,which may increase PPAR-γ and GLUT-4 expression levels,
