摘  要:目的比较Bliss法中Probit、Logit、Gompit和Angle 4种模型计算半数有效量、半数致死量及治疗指数的差异,探究更优的计算模型。方法通过统计8组不同药物的ED50及LD50实验结果,运用Regu Stats1.0软件计算出ED50、LD50估计值、置信限、标准误和治疗指数。运用SPSS19.0软件对估计值及标准误进行正态分布检验,并进行配对t检验。结果对于ED50及LD50的计算Gompit模型与其他三种模型的估计值存在统计学意义上的差异,4种模型中Angle模型标准误最小,Probit模型次之。对于治疗指数的计算四种模型间无统计学意义上的差异。结论对于半数有效量及半数致死量的计算,推荐选择Angle模型和Probit模型。对于治疗指数的计算,选择四种模型均可。实际应用中应结合不同模型的适用条件加以选择。Objective To compare the differences of the Bliss four models( Probit,Logit,Gompit and Angle model) in calculating the median effective dose,the median lethal dose and the therapeutic index,and try to find a superior model. Methods Through the statistics of eight different groups of drugs ED50 and LD50experimental results,using Regu Stats1. 0 software to calculate ED50,LD50 estimated values,standard errors and confidence limits,calculated the therapeutic index,analyzed the estimated value and standard error by normal distribution test and paired t test. Results Gompit model is statistically significant different from other three models in calculating the estimated ED50 and LD50values. Among the four models,the standard error of Angle model is minimal,followed by Probit model. In calculating the therapeutic index,there are no statistically significant differences among the four models. Conclusions For the calculation of median effective dose and median lethal dose,Angle model and Probit model are recommended. For the calculation of the therapeutic index,all the four models can be used. In practical applications,the applicable conditions of different models should be considered in order to choose the most suitable model.
