摘 要:目的检测钙黏蛋白家族成员(CDHR)3、白细胞介素(IL)-33、DNA修复蛋白(RAD)50基因位点多态性与儿童支气管哮喘的易感性。方法选择2012年12月至2014年6月于青岛大学附属医院儿科支气管哮喘门诊就诊及住院治疗的100例支气管哮喘患儿为研究对象,并将其纳入支气管哮喘组(n=100)。选择同期于本院儿科门诊体检中心体检的100例健康儿童纳入对照组(n=100)。本研究遵循的程序符合青岛大学附属医院人体试验委员会所制定的伦理学标准,得到该委员会批准,分组征得受试对象监护人的知情同意,并与之签署临床研究知情同意书。两组儿童年龄、性别构成比、民族等基本资料比较,差异均无统计学意义(P>0.05)。采用质谱单核苷酸多态性(SNP)技术,对两组儿童CDHR3、IL-33、RAD50基因进行分析。结果两组CDHR3、IL-33、RAD50基因位点基因型分布均符合Hardy-Weinburg平衡定律。两组CDHR3基因rs6967330位点和IL-33基因rs928413位点基因型与等位基因频率比较,差异无统计学意义(χ2=3.442,3.071,2.895,2.829;P=0.179,0.088,0.235,0.124)。两组RAD50rs6871536位点基因型与等位基因频率比较,差异均有统计学意义(χ2=6.635,9.200;P=0.036,0.003)。结论 CDHR3基因rs 6967330位点多态性和IL-33基因rs928413位点的多态性可能与儿童支气管哮喘无相关性。RAD50基因rs6871536位点多态性可能与儿童支气管哮喘有相关关系。Objective To evaluate the potential relationship between polymorphisms of cadherinrelated family member(CDHR)3,interleukin(IL)-33 and DNA repair protein(RAD)50and bronchial asthma.Methods From December 2012 to June 2014,a total of 100 children with bronchial asthma who were diagnosed and treated at the Affiliated Hospital of Qingdao University were included in the study as bronchial asthma group(n=100).Meanwhile,100 healthy volunteers were recruited for this study as control group(n=100).The CDHR3,IL-33,RAD50 gene were analyzed by single nucleotide polymorphisms technique.The study protocol was approved by the Ethical Review Board of Investigation in Human Being of Affiliated Hospital of Qingdao University.Informed consents were obtained from each participants′parents.Results The alleles of CDHR3,IL-33,RAD50 gene were in Hardy-Weinburg balance.There were no significant differences in rs6967330 of CDHR3 and rs928413 of IL 33 gene polymorphisms between two groups(χ^2=3.442,3.071,2.895,2.829;P=0.179,0.088,0.235,0.124).Significant differences were found in rs6871536 of RAD50 gene polymorphisms between two groups(χ^2=6.635,9.200;P=0.036,0.003).Conclusions The rs6967330 and rs 928413 polymorphisms were not correlated with asthmatic susceptibility,rs6871536 polymorphisms may be the susceptible genotype of bronchial asthma in children.
