摘　　要：目的观察硫氧环蛋白过氧化物酶3(Prdx-3)在血管紧张素Ⅱ(AngⅡ)诱导心肌细胞肥大中的表达。方法体外培养心肌细胞(H9C2)随机分为对照组和AngⅡ组(AngⅡ1×10^(-6)mol/L处理)。Real time PCR法检测脑钠素(BNP)mRNA表达,二氯荧光素二乙酸(DCFH-DA)检测活性氧(ROS)水平,Western blot法检测Prdx-3蛋白表达。结果①与对照组比较,AngⅡ刺激6、12、24、48 h的BNP mRNA分别增长(1.10±0.08)、(1.30±0.18)、(1.80±0.25)、(2.00±0.37)倍,除刺激6 h外,其他时间点的差异均有统计学意义(P<0.05)。②AngⅡ刺激5、10、20、40 min和1 h ROS水平分别是(3740±75)、(3911±91)、(4330±143)、(4073±335),(3879±226),与对照组(3426±197)比较,刺激5、10、20 min时差异有统计学意义(P<0.05)。③AngⅡ刺激30 min、1 h、3 h和6 h时Prx-3蛋白的表达值分别为(0.72±0.11)、(0.50±0.07)、(0.42±0.08)和(0.35±0.08),与对照组(0.33±0.05)比较,AngⅡ刺激30 min和1 h时差异均有统计学意义(P<0.05)。结论在AngⅡ诱导心肌细胞肥大过程中伴随着Prdx-3表达的上调,推测Prdx-3表达与降低ROS有关。Objective To observe the expression of peroxiredoxin-3 （Prdx-3） in AngiotensinII （AngII） inducing car-diomyocytes hypertrophy. Methods Cultured cardiomyocytes were randomly divided into control group and Ang IIgroups （dealed with Ang II 1＆#237;10-6 mol/L）. BNP mRNA and reactive oxygen species （ROS） were measured by Real-time PCR and DCFH-DA;while Prdx-3 expression was by Western blot. Results①Compared with control group, BNP mRNA expression increased （1.10±0.08）, （1.30±0.18）, （1.80±0.25）, （2.00±0.37） times respectively when Ang II stimu-lated at 6, 12, 24, 48 h, except at 6 h, the differences were statistically significant （P〈 0.05）. ②The level of ROS in control group was （3426±197）, ROS increased to （3740±75）, （3911±91）, （4330±143）, （4073±335）, （3879±226） respec-tively when Ang II stimulated at 5, 10, 20, 40 min and 1 h, compared with control group, the differences were statisti-cally significant （P〈 0.05）. ③The level of Prdx-3 in control group was （0.33±0.05）, Prdx-3 increased to （0.72±0.11）, （0.50±0.07）, （0.42±0.08）, （0.35±0.08） when AngIIstimulated at 30 min, 1 h, 3 h and 6 h, compared with control group, the differences of Ang II stimulated at 30 min, 1 h were statistically significant （P 〈 0.05）. Conclusion Cardiomy-ocytes hypertrophy is associated with Prdx-3 upregulation induced by AngiontensinⅡ.
