摘 要:目的探讨肾阳虚简易有效的建模方法和对甘氨酸受体(GlycR)mRNA表达的影响。方法用腺嘌呤皮下注射构建肾阳虚大鼠模型,观察大鼠行为变化,测定其自主活动度、脏器指数和体重,做肾脏切片,并运用Real-time RT-PCR测定中枢神经GlycR mRNA的相对表达量。结果造模后,大鼠肾脏、睾丸指数增大;肾皮质固缩,与髓质界限模糊,有灰褐色沉积物;与正常组相比,大鼠自主活动度降低(P<0.05),GlycR mRNA的相对表达明显增高(P<0.05)。结论用腺嘌呤建立肾阳虚大鼠模型方法可靠方便,可能会增强GlycR mRNA的表达。Objective To explore a simple and effective method of Kidney- Yang deficiency in rats,and the expression of Glycine receptor( GlycR) mRNA. Methods Rats were injected subcutaneously with adenine to build Kidney- Yang deficiency rats model. The behavior,kidney,testis mass,kidney biopsy and body weight of Kidney- Yang deficiency rats were measured and compared to rats without any treatments( negative control group). Real- time RT- PCR was employed to measure GlycR mRNA expression. Results Kidney index and testis index increased in Kidney- Yang deficiency rats. Renal cortical shrinkage and have gray- brown sediment,the border of cortex and medulla is unclear. Compared to Kidney- Yang deficiency rats,locomotor activity in low- dose group of rats significantly reduced( P 0. 05),and GABAa mRNA expression significantly increased( P 0.05). Compared to the negative control group,GlycR mRNA of rats was significantly increased( P 0. 05). Conclusion It is available for using adenine to build a Kidney- Yang deficiency rat model and Kidney- Yang deficiency rats have increased glycine receptor expression.
