摘  要:目的建立肝郁脾虚型肠易激综合征(IBS)模型组和对照组结肠组织蛋白质双向电泳图谱,结合质谱技术鉴定并分析差异表达蛋白,对候选表达差异蛋白进行蛋白质功能和相关研究,进一步阐明肝郁脾虚型IBS病理机制。方法采用双向凝胶电泳法分离IBS模型组和正常对照组结肠粘膜差异蛋白,运用MALDI TOF/TOF串联飞行时间质谱仪进行鉴定,运用Gene Ontology和Pathway Studio生物信息学方法将所鉴定的差异蛋白进行功能分类,分析IBS病理机制中关键蛋白。结果首次运用蛋白组学的方法寻找IBS模型组和正常对照组结肠的差异表达蛋白特征谱,结果发现差异倍数在2.5倍以上的差异蛋白有15个,其中2个蛋白表达上调,13个蛋白表达下调。结论热休克蛋白、硫氧还蛋白、氯离子通道蛋白等可能是肝郁脾虚型IBS病理机制中潜在分子标志物。Objective To establish the two- dimensional gel electrophoresis of colonic tissue proteins of irritable bowel syndrome( IBS) model group and control group,combined with the mass spectrometry identification and analysis of differential expression proteins,and clarify the pathomechanism of IBS from studying the function of the candidate proteins. Methods The compositions of the differential proteins of colonic mucosa from the IBS model group and control group were determined by two- dimensional gel electrophoresis,identified by MALDI TOF / TOF and time- of- flight mass spectrometer. The functions of differential protein were classified by Gene Ontology and Pathway Studio bioinformatics,analysing the key proteins of IBS. Results The research has been studied for the first time by proteomics to find the character spectrum of differential expression protein from the IBS model group and control group. The result showed the differential proteins which differences in 2. 5 times or more were 15. Conclusion Heat shock protein,Thioredoxin and Chloride Channel Protein may be the potential molecular markers of IBS pathomechanism.
