摘  要:【目的】分析DNA条形码技术用于鉴定新疆蝗蝻的可行性,同时对这些样本进行了系统发育分析。【方法】利用目前广泛应用的DNA条形码识别技术,以新疆地区的蝗蝻为靶标扩增其线粒体DNA细胞色素C氧化酶亚基I(mitochondrial cytochrome coxidase subunit I gene,mtD NA COI)基因(658 bp),联合运用相似性方法 (BLAST in NCBI and species identification tool at BOLD Systems v3)和NJ聚类分析对所获得的蝗蝻碱基序列进行物种鉴定,系统发育分析则采用ML建树法。【结果】研究表明所采集到的蝗蝻均能准确确定其分类地位;在系统发育分析中,如果按照国际通用OSF分类体系,Oedipodinae则为一单系群,而按夏氏分类体系,研究支持将原本属于斑翅蝗亚科的小车蝗属归为飞蝗亚科,有关的系统发育关系如下:(((飞蝗属+车蝗属)+小车蝗)+斑翅蝗属)。然而,按夏氏分类体系,网翅蝗亚科未能形成一单系群,其下五属之间的系统发育关系为:(土库曼蝗属+((草地蝗属+异爪蝗属)(戟纹蝗属+米纹蝗属)))。【结论】由此推断基于COI基因的DNA条形码技术及综合运用相似性方法和NJ聚类分析可以用于蝗蝻的快速准确鉴别,该方法在蝗虫早期防治过程的种类鉴定中具有重要的应用价值。在系统发育分析中,658 bp的COI序列虽能解决部分系统发育问题,然而,丰富的样本量及包含更多系统发育信息的基因片段或数据集则有助于提供更全面正确的系统发育关系。【Objective】This research is to check the utility of DNA barcoding for species identifl cation of nymph grasshoppers from Xinjiang by using 658 bp sequence of mitochondrial gene cytochrome c oxidase subunit I. Meanwhile,phylogenetic relationships of these Caelifera species were analyzed using maximum likelihood( ML) based on the short sequence.【Method】Similarity methods( BLAST and species identification tool at BOLD website) and NJ( Neighbour Joining) clustering analysis were used in combination for species identifi cation,and maximum likelihood( ML) was adopted in phylogenetic analysis.【Result】All nymphs were successfully discriminated to either species or genus level. The phylogenetic analysis indicated that Oedipodinae in OSF system was a monophyletic group and the ML result supported the assignment of Oedaleus to Locustinae instead of Oedipodinae in Xia's system. The relationships among the four genera were((( Locusta + Gastrimargus)+ Oedaleus) + Oedipoda). However,the Arcypterinae in Xia's system could not form the monophyletic group. The phylogeny of Arcypterinae five genera were( Ramburiella +(( Stenobothrus + Euchorthippus)( Dociostaurus + Notostaurus))).【Conclusion】Our study showed that the fragment of658 bp of the first half of the mitochondrial cytochrome C oxidase subunit I gene( COI) could be used as DNA barcoding to provide a powerful foundation for accurate and unambiguous identification of nymph grasshoppers,allowing the development of efficient pest control strategies,and improving understanding larva species diversity,phylogenetic patterns,and evolutionary processes and ecosystem research. Furthermore,we also suggested that the NJ clustering analysis and similarity methods are a good choice for DNA barcoding analysis. Basically,the phylogeny constructed by the short 658 bp sequence can provide some useful phylogenetic information,however,more taxa and long sequence or dataset with more phylogenetic signals,for example
