摘  要:目的检测宫颈癌患者鳞癌组织中CXCR4的表达,观察其与宫颈鳞癌各种临床病理参数的关系,从而为探讨CXCR4与宫颈癌侵袭性的机制研究提供一定的实验依据。方法收集我院妇产科收治的145例宫颈癌手术治疗患者临床病理资料及切除标本,采用免疫组织化学SP法检测标本中CXCR4的表达情况,并通过Transwell实验了解CXCR4过表达的细胞迁移情况。结果不同FIGO病理分期、不同组织分化程度及不同淋巴结转移情况的患者之间,CXCR4表达的阳性率差异有统计学意义(P<0.05),FIGO分期为Ⅱ期及Ⅲ期者阳性率较高,组织分化程度越低者阳性率越高,发生淋巴结转移者CXCR4表达的阳性率高于未转移者。宫颈鳞癌细胞迁移实验结果显示,除CXCR4过表达3 h和6 h两个时点外,其余各时间点宫颈鳞癌细胞过表达CXCR4组与对照组相比较,平均迁移细胞数的差异均具有统计学意义(P<0.05)。结论宫颈鳞癌组织中CXCR4高表达可能在宫颈鳞癌的侵袭、转移中起重要作用,对细胞侵袭和转移能力有促进作用。Objective To explore the expression status of CXCR4 protein in cervical cancer and investigate the clinical utilization of both CXCL12 and CXCR4 and the underlying mechanism. Methods Immunohistochemistry using SP was used to detect expression of CXCL12 and CXCR4 in 120 samples of human cervical cancer and the matched control. The correlations between expressions of CXCL12, CXCR4 and clinical pathological parameters for cervical cancer were analyzed respectively. To determine whether CXCR4 participated into the metastasis of cervical cancer, we employed transwell assays. Results CXCL12 and CXCR4 were detected in both cervical carcinoma tissues and normal tissues. However, compared to the normal tissues, the expression levels of CXCL12 and CXCR4 were both up-regulated in cervical carcinoma tissues(CXCR4: χ2=38.34, P〈0.05; CXCL12: χ2=16.61, P〈0.05). The expression of CXCL12 and CXCR4 were significantly different(P〈0.05). The expression levels of CXCL12 were closely correlated with pathologic grading, FIGO, and bone metastasis(P〈0.05). CXCR4 were closely correlated with pathologic grading, FIGO, and bone metastasis(P〈0.05). CXCR4 overexpression promote cervical cancer metastasis. Conclusion CXCR4 and CXCL12 are correlated with the biologic behavior and prognosis of cervical carcinoma. CXCL12 and CXCR4 are helpful to predict metastasis of cervical carcinoma.
