硝普钠对保存期间悬浮红细胞变形性的作用研究

摘  要:目的探讨硝普钠对保存期间悬浮红细胞变形性的影响。方法来源于血站的50袋悬浮红细胞,200mL/袋,在保存期的第1、3、5、7、14、21、28、35天(D1、D3、D5、D7、D14、D21、D28、D35)共8个时间点,各取20mL,分成4管(5mL/管),其中,1管不做处理为对照组(A组),另外3管分别添加硝普钠,配置成终浓度为B组1μmol/L、C组5μmol/L和D组10μmol/L。检测4组的红细胞变形指数、聚集指数和刚性指数。结果与A组红细胞变形指数比较,B组和D组差异无统计学意义(P>0.05),C组明显降低(P<0.05);与A组聚集指数和刚性指数比较,B、C、D 3组之间差异无统计学意义(P>0.05)。结论浓度为5μmol/L的硝普钠能够改善悬浮红细胞的变形指数,但对聚集指数和刚性指数无明显影响。Objective To investigate the effects of sodium nitroprussiate on deformability of suspension red blood cells during the storage period.Methods Fifty bags of blood samples(200mL/bag)were collected from blood center.20 mL of blood samples was fractionated to four tubes(5mL/tube)during incubation of blood samples.Three tubes of blood samples has been incubated with different concentrations of sodium nitroprussiate(SNP)at 8-phase(D1,D3,D5,D7,D14,D21,D28,D35)of storage.Concentration of SNP were 1μmol/L(group B),5μmol/L(group C)and 10μmol/L(group D).The final tube is control group(group A)without treatment.The relevant indicators of erythrocyte deformability(deformability index,aggregation index,stiffness index)have been detected after incubation.Results Compared to group A,deformability index of groups B and D had no significant changes in storage periods(P〉0.05),but deformability index of group C were gradually reduced(P〈0.05),aggregation index and stiffness index had no significant changes in groups B,C and D(P〉0.05).Conclusion When the concentration of SNP were 5μmol/L,SNP was useful to improve the deformability index of suspension red blood cells,but there were no effects on aggregation index and stiffness index.
