摘 要:目的采用响应面法对白簕叶总黄酮的回流提取工艺进行优化。方法通过单因素试验选取因素和水平;以白簕叶总黄酮质量分数为指标,选取乙醇体积分数、提取时间和料液比3个因素进行BoxBehnken中心组合设计。结果白簕叶总黄酮的最佳提取工艺条件为:提取时间为76 min,乙醇体积分数为49%,液料比为42∶1(m L∶g)。在此条件下,白簕叶总黄酮的质量分数达5.12%。结论响应面法对白簕叶总黄酮的回流提取方法有较好的预测性,可为白簕叶总黄酮的高效提取提供参考。Objective To investigate the reflux extraction conditions of total flavonoids from Acanthopanax trifoliatus( L.) Merr. leaves by surface response methodology. Methods On the basis of single factor experiments,the concentration of total falvonoids was set as response value,and the surface response methodology was used to optimize extraction of total flavonoids from Acanthopanax trifoliatus( L.) Merr.leaves with three factors which were ethanol concentration,extraction time,and liquid-solid ratio. Results The optimal extraction condition was as follows: the ethanol concentration was 49%,the extraction time was76 min,the liquid-solid ratio was 42∶ 1( m L∶ g),and the average total flavonoid concentration was 5. 12%.Conclusions Surface response methodology has good predictability on the optimization of reflux extraction of total flavonoids from Acanthopanax trifoliatus( L.) Merr. leaves,and the optimal extraction condition can provide reference for high-performance extraction of the total flavonoids.
