摘  要:目的利用正交设计法优选火炭母中总黄酮的提取工艺。方法以芦丁为对照,硝酸铝为显色剂,用紫外分光光度法在510 nm处测定总黄酮的含量。采用L9(34)正交实验设计,以火炭母中总黄酮含量为考察指标,考察乙醇浓度、料液比、提取时间、提取温度4个因素对提取的影响。结果最佳提取工艺为乙醇浓度为70%,料液比1∶40,提取时间为1.5 h,提取温度95℃。结论优选工艺简单、易行、经试验重复性好,为进一步的工艺开发提供了较好的参考价值。Objective To optimize extraction technology of total flavonoids in Polygonum chinensis L.. Methods The ultraviolet spectrophotometry method was used to measure the total flavonoids content in ethanol extract of Polygonum chinensis L. with different polar solvent extraction, with aluminium nitrate as color reagent and the rutin as standard substance and the maximum absorption wave-length of 510 nm. By L9(34) orthogonal experimental design, with Polygonum chinensis L., as index of total flavonoids were investigated ethanol concentration, ethanol dosage, extraction time and extraction temperature four factors on the extraction. Results The optimum extraction process was 70 % ethanol, 70 times the amount of reflux extraction, 2 h and 95 ℃. Conclusion The experimental reproducibility of the process, the process of further development of a better reference value.
