摘  要:目的观察灵芝孢子粉对镉致雄性大鼠睾丸雄激素结合蛋白和抑制素的影响。方法 45只SD雄鼠,随机均分为3组:对照组、Cd组、灵芝孢子粉拮抗组(GLS+Cd)。灵芝孢子粉拮抗组用5mg/kg灵芝孢子粉灌胃,对照组与Cd组灌胃等量生理盐水,每天1次。Cd组和GLS+Cd组腹腔注射Cd Cl22mg/kg,隔天1次。对照组腹腔注射等量生理盐水。3个月后分3批处死,间隔为1个月。用RT-PCR法对抑制素和雄激素结合蛋白的表达量进行定量分析,睾酮定量分析酶联免疫检测试剂盒测定血清睾酮浓度。结果在镉染毒1个月时,Cd组ABP(雄激素结合蛋白)表达量应激性升高,在第3个月Cd组ABP表达量低于灵芝孢子粉拮抗组,有统计学意义(P<0.05);随着镉染毒时间延长,第2、3个月Cd组INH-B(抑制素B)表达量低于灵芝孢子粉+Cd组,有统计学意义(P<0.05);镉染毒3个月时,与对照组比较,Cd组和灵芝孢子粉拮抗组血清睾酮水平明显降低(P<0.05);与Cd组比较,灵芝孢子粉+Cd组血清睾酮水平浓度高(P<0.05)。结论灵芝孢子粉可以通过提高ABP、INH-B的表达对镉致雄性大鼠支持细胞损伤有一定的保护作用。Objective To investigate the protective effects of ganoderma lucidum spore(GLS) on androgen protein(ABP) and inhibin(INH) expression by cadmium(Cd) in rat testis.Methods Forty five male SD rats were randomly and equally divided into blank control group,Cd group,GLS + Cd group.5 mg / kg of GLS was given to GLS anti-cadmium group every day,the control group and Cd group were given with 0.9% normal saline,the cadmium alone group and GLS anti-cadmium group were intraperitoneally injected with Cd Cl2 solution of 2mg / kg,every other day,for three months.Then the rats were sacrificed after 30,60 and 90 days respectively and their blood and testes were reserved for further tests.The mRNA of androgen protein(ABP) and inhibin-B(INH-B) were measured by RT-PCR,and the quantitative analysis of testosterone(T) was determined by the ELISA method.Results Compared with GLS + Cd group,the increase of ABP in Cd group after 1 months,and the decrease of ABP in Cd group after 3 months were significant(P〈 0.05).Compared with blank and GLS+ Cd group,the INH-B expressions decreased in Cd group with the time(P〈 0.05) after 2 or 3 months.Meanwhile,the decrease of T in Cd and GLS + Cd group was significant(P 〈0.05) after 3 months compared with control group,and the decrease of T in Cd group was more significant(P〈 0.05) after 3 months than that of GLS + Cd group.Conclusion Ganoderma lucidum spore could significantly improve the expressions of ABP and INH-B,so as to achieve the role of protecting Sertolicells in rat testis induced by Cd.
