黄山花楸成熟合子胚的萌发及愈伤组织的诱导

摘　　要：以黄山花楸成熟胚为材料在MS基本培养基上萌发,在不同激素组合的诱导培养基上诱导愈伤组织,统计萌发率及诱导率,结果表明去掉种皮剥离出的成熟胚在MS培养基上均可萌发,提示黄山花楸种子萌发的障碍因素是种皮的物理束缚;诱导愈伤培养基添加6-BA和NAA均可诱导黄山花楸合子胚产生愈伤组织,但愈伤出愈率存在差别,最佳愈伤诱导培养基为MS+6-BA1.0mg/L+NAA0.1mg/L,愈伤组织的诱导率达100%。The mature embryos of Sorbus amabilis were cultured for germination on MS medium. The mature embryos of Sorbus amabilis were cultured for callus on MS medium.supplemented with different hormone combinations.Counting for the germination rate and induction rate. Experimental results show that all mature zygotic embryos without seed coats can germinate,it is proved that physiologica dormancy does not exsit in Sorbus amabilis seeds and physical dormancy is main lycaused by mechanical obstruction.The mature embryos of Sorbus amabilis were cultured for callus on MS medium.supplemented with 6-BA and NAA all could be induced, but there were obvious different.The best induced medium were MS ＋6-BA1.0mg/L＋NAA0.1mg/L, induction rate was 100%.
