摘　　要：目的 研究抑制去泛素化酶USP14的活性对大鼠心肌成纤维细胞（CFs）增殖的影响.方法 用胰酶消化法和差速离心法分离培养新生SD大鼠CFs,加入不同浓度去泛素化酶USP14的抑制剂IU1作用48小时后,采用MTS、细胞计数法和结晶紫染色法检测CFs的增殖,采用LDH释放检测测定IU1对CFs的细胞毒性作用.结果 IU1可以浓度依赖的抑制CFs的增殖,并且没有明显的细胞毒性作用.结论 抑制去泛素化酶USP14活性可以抑制CFs的增殖,去泛素化酶USP14可能是一个抗心肌纤维化的重要靶点.Objective To evaluate the effect of inhibiting deubiquitinase USP14 activity on the proliferation of cultured rat cardiac fibroblasts （CFs）.Methods Cultured CFs from neonatal SD rats were isolated by trypsinization and differential centrifugation,treated with various dose of IU1,the inhibitor of deubiquitinase USP14,and examined after 48 h.The proliferations of CFs were detected by MTS,cell counts and crystal violet staining,the CFs cellular toxicity of IU1 was determined by LDH release detection.Result IU1 could inhibit the proliferation of CFs in a concentration-dependent manner and had no significant cytotoxicity.Conclusion The inhibitory activity of deubiquitinase USP14 can suppress the proliferation of CFs,deubiquitinase USP 14 may be an important target of anti-cardiac fibrosis.
