摘  要:目的:探讨不同浓度异丙肾上腺素在体外对婴儿血管瘤内皮细胞生长的影响及β-肾上腺素受体在婴儿血管瘤发病机制中的作用。方法将增殖期婴儿血管瘤内皮细胞分为空白组(不含药物培养基)及异丙肾上腺素低浓度组(5、10、20 mg/L)与高浓度组(50、100、200 mg/L)。药物作用1~10 d 采用 MTT 法分别测定各组细胞的生长曲线。结果低浓度组24~96 h 对细胞生长呈现刺激作用;96 h 时,低浓度组与空白对照组的吸光度值差异有统计学意义(P <0.05)。而高浓度组在24~96 h 对细胞生长呈现抑制作用,与空白对照相比,高浓度组细胞的吸光度值差异有统计学意义(P <0.05),并且24~96 h 浓度越高,作用越强。结论β-肾上腺素受体在婴儿血管瘤发病中有重要作用;异丙肾上腺素对于体外培养的增殖期血管瘤内皮细胞的刺激作用与浓度相关。Objective To investigate the different concentrations of isoproterenol in vitro effects on infant hemangioma endothelial cell growth and to study the function of beta adrenergic receptors in the pathogen-esis of infantile hemangioma.Methods Infantile hemangioma endothelial cells in proliferating phase were divided into three groups,the blank group (without drug culture medium),low concentration group(5, 10,20 mg/L)of isoproterenol and high concentration group (50,100,200 mg/L).The growth curve of cells in each group were measured after a certain time by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT)method.Results Low concentration group showed stimulation affection on cell growth within 24-96 hours;and in the high concentration group,cell growth were inhibited within 24-96 hours,and the hiher concentration,the stronger the effect within 24-96 hours.Conclusion Beta ad-renergic receptors play an important role in the pathogenesis of infantile hemangioma;stimulation effection of isoproterenol is related with concentration of drug for in vitro culture of hemangioma endothelial cells.
