摘 要:目的建立高效液相色谱(HPLC)法测定新伤止痛颗粒中盐酸小檗碱含量。方法采用反相HPLC法,色谱柱为phenomenex Gamini-C18柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.033 mo L/L磷酸氢二钾溶液(38∶62),检测波长为345 nm,流速1.0 m L/min,柱温35℃。结果盐酸小檗碱进样量在0.01184~0.1776μg范围内与峰面积呈良好的线性关系(r=0.99996),平均加样回收率为98.01%,RSD=1.83%(n=6)。结论该方法操作简单、准确、重现性好,可用于新伤止痛颗粒中盐酸小檗碱的含量测定。Objective: To establish a method for determining berberine hydrochloride content in Xinshangzhitong Granule by HPLC. Methods: Berberine hydrochloride was analyzed by using Phenomenex Gamini-C18column(250 mm×4.6 mm,5 μm) with the mobile phase of acetonitrile-0.033 mol/L dipotassium hydrogen phosphate(38∶62) at the flow rate was 1.0 m L/min. The detection wavelength was 345 nm and the column temperature was 35℃. Results: The linear of berberine hydrochloride was in the range of 0.01184~0.1776 μg(r=0.99996). The average recoveries were 98.01% with RSD of 1.83%(n=6). Conclusion: The method is simple,reliable and accurate. It can be used for the determination of berberine hydrochloride content in Xinshangzhitong Granule.
