摘 要:为探索培养基对脂肽种类及产率的影响,本文考察了Bacillus subtilis XF-1在5种培养基中发酵的生物量和丰源素、表面活性素及伊枯草菌素等环脂肽化合物产率,并通过HPLC分析了发酵液中化合物种类的变化。试验结果表明,各培养基在相同条件下生物量由大到小的顺序为MOLP(2.07 g/L)、PSA(1.28 g/L)、LB(1.03 g/L)、Landy(0.326 g/L)、蔗糖取代葡萄糖的Landy培养基(LS,0.375 g/L)。经过酸沉醇提取后获得脂肽混合物,其质量由大到小的顺序为:MOLP(0.351 g/L)、PSA(0.176 g/L)、LB(0.201 g/L)、Landy(0.501 g/L)、LS(0.517 g/L)。最适合菌体生长的是MOLP培养基,脂肽产率最高的是LS,其次是Landy培养基。5种培养基发酵液中所含环脂肽类化合物色谱图基本一致。LS比Landy提高菌体生物量15%,提高脂肽类抗生素3%,因此Landy培养基和LS培养基可作为XF-1脂肽化合物合成专用培养基。Bacillus subtilis XF- 1 was cultured in 5 media to investigate the changes of its biomass,production ability of cyclic lipopeptides family( CLPs) such as fengycin,surfactin and iturin,and the supernatants were extracted with acid and precipitated with alcohol,and then the precipitants were detected by HPLC respectively. The biomass were 2. 07,1. 28,1. 03,0. 326 and 0. 375 g /L and the mass of CLPs were 0. 351,0. 176,0. 201,0. 501 and 0. 517 g / L in MOLP,PSA,LB,Landy and LS media,and all of the HPLC chromatograms are very similar. MOLP was the best growth medium,and LS medium derived from Landy whose glucrose is replaced with sucrose produced 15% more bacteria and 3% more CLPs than Landy did. Therefore,Landy and LS might be used as specific media for XF- 1 to produce CLPs.
