摘  要:对一种新开发出的酸性蛋白酶H(简称酸性酶H)的酶学性能及其在皮革软化中的应用进行了研究。试验结果表明:该酶在50℃,pH值3时酶活力较高。37℃时酶释放活力的最佳时间为60min内。当Cr^(3+)浓度超过2%、NaCl浓度大于1%时,酶活力呈明显下降趋势。应用试验结果表明:酸性酶H用量为0.8%、温度37℃、浸酸软化裸皮90min后,绵羊皮的柔软度及弹性增加。组织学研究表明:经酸性酶H软化后的粒面层纤维呈现蓬松状且空隙增多,粒面层结构完整。网状层纤维束分散明显,纤维间隙增大。猪蓝湿革经0.8%酸性酶H、温度45℃软化后,厚度减少和面积增加显著,折痕减轻明显,粒面完好。因此酸性酶H经过进一步改良之后,有望在制革生产中得以应用。The enzymology properties and application effects on leather bating of a new acid proleatytic enzyme which called acid enzyme H was studied. The results show that the acid enzyme H has a higher enzyme activity at 50% of pH 3. The optimum time is 60min for releasing enzyme activity at 37℃. The enzyme activity has trend down when the concentration of Cr3+ is over 2% and NaCI is over 1%. The application results show that the dosageof enzyme H is 0. 8% at 37℃ for 90min when it used in bating and pickling of sheepskin, the softness and flexibility of sheepskin are improved. The histological observations show that after bat- ing, the collagen fibers of grain layer became loose and the space of fiber increases evidently, while the structure is integrated. The fiber bundles of reticular layer disperse obviously and the space increases. The wet blue are bated by 0. 8% of enzyme H at 45%. The thickness of wet blue decreases and area increases, the crease significantly reduces, pores are clearly visible. Thus acid enzyme H can be expected to be used in leather making after further improvement.
