不同方法所制备黄芪多糖含药血清对神经干细胞增殖分化的影响

摘　　要：【目的】观察不同给药方法制取的APS含药血清对神经干细胞增殖分化的影响。【方法】用连续注射法、一次性大剂量注射法和灌胃法制取APS含药血清,采用间接免疫荧光法检测Nestin,β-tubulinⅢ和GFAP,通过CCK-8 Brdu检测细胞增殖,Elisa检测VEGF、NGF含量,Western Blot法检测PI3K、P-Akt、Casepase-9蛋白表达。【结果】APS含药血清组与对照组在增殖分化中有明显差异(P<0.05),可提高VEGF、NGF含量,提高PI3K、P-Akt蛋白表达,降低Casepase-9蛋白表达。其中APS腹腔注射含药血清组优于APS灌胃含药血清组,而连续注射法与一次性大剂量注射法含药血清组无明显差异(P>0.05)。【结论】APS含药血清组可能通过调节VEGF、NGF含量,活化PI3K/Akt信号通路,促进神经干细胞增殖分化,Akt磷酸化后,调节Casepase-9,延缓细胞凋亡。效果与其给药方式有关,腹腔注射法优于灌胃法,但与腹腔注射给药时间无关。[Objective] To observe the effect of APS drug serum in different administration methods proliferation and differentiation of NSCs.[Methods] By continuous injection method,single dose injection and gavage to get the APS drug serum,we detected the expression of Nestin,β-tubulin Ⅲ and GFAP by indirect immunofluoresence.The proliferation of NSCs was evaluated by CCK-8 Brdu.The levels of NGF and VEGF were detected by Elisa.The expression of PI3K,P-Akt,Casepase-9 protein was detected by Western blot.[Results] There was obvious difference in cell proliferation of APS drug serum group and the control group （P ＜ 0.05）,APS drug serum group can increase the expression level of NGF、VEGF、PI3K、P-Akt,reduce the expression of Casepase-9 protein.in which intraperitoneal injection of APS drug serum group was better than that of gavage APS drug serum group,and continuous injection method and single dose injection drug serum group no significant difference （P ＞ 0.05）.[Conclusion] APS drug serum can promote the proliferation and differentiation of neural stem cells,by regulate the content of NGF,VEGF in blood,the activation of PI3K/Akt signal pathway,promote Akt phosphorylation,accommodate the Casepase-9,to delay of cell apoptosis.With the relevant modes of administration,intraperitoneal injection is better than that of gastric lavage,but with intraperitoneal injection time independent.
