表皮干细胞中CK15和CD49f阳性率与年龄关系的初步探究

摘　　要：目的:探究人体包皮组织中分离的表皮干细胞CK15和CD49f阳性表达率与年龄的关系。方法:利用双酶法从1、11、28、31岁4个年龄组皮肤组织中分离表皮干细胞,用CK15和CD49分别进行标记,用流式细胞仪检测阳性率。结果:在研究的4个年龄组中,11岁年龄组皮肤中的细胞CK15和CD49标记物阳性率最高,分别为70.19%和25.27%;流式细胞仪分选出的CK15阳性和CD49f阳性表皮干细胞在体外可呈现克隆生长。结论:11岁年龄组的表皮干细胞活性较强,更适宜组织工程种子细胞的选用。Objective： To investigate the relationship between age and the positive expression of CK15 and CD49 for epidermal stem cells.Methods： Epidermis was obtained from different tissue of group aged 1,group aged 11,group aged 28 and group age 31.They were separated and cultured by digesting skin for several times with dis-pase Ⅱ and trypsin-EDTA.The epidermal stem cells were marked with CK15 and CD49 f and sorted by flow cy-tometry.Results： In aged 11 group,the epidermal stem cells positive expression for CD49 and CK15 were morehigher than other groups.The positive expression rate of CD49 and CK15 were 70.19% and 25.27% in aged 11 group.Epidermal stem cells sorted by flow cytometry were in good growth status and formed clone group.Conclusion： Epidermal stem cells obtained from group aged 11 showed better activity and more suitable for tissue engi-neering seed cells than other groups.
