摘  要:目的 对块根糙苏进行生药研究并测定糙苏中山栀苷甲酯的含量,并对糙苏药材的质量标准提供依据.方法 从植物学角度及应用显微镜对块根糙苏进行研究.采用高效液相色谱法测定山栀苷甲酯的含量:以70%甲醇超声(功率300 W,频率40 kHz)处理糙苏药材,Waters SunFireTM C18柱(250 mm ×4.6 mm,5 μm);以乙腈-水(9∶91)为流动相;检测波长:236 nm;流速:1.0 ml/min;柱温:25℃;进样体积:10μl.结果 块根糙苏的药材有块根和细根相连,形状较有特色;根横切面特征表明,木质部导管较大,且有明显的裂隙.粉末特征网纹导管特别多,而且大小形状不一,木纤维长梭形等特征可做本药材的鉴别依据.以峰面积和对照品量作线性回归,回归方程为(Y=1 464 093.767X-8 682.4,r=0.999 7),山栀苷甲酯在0.065 26~0.900 6 ng内成良好线性关系,平均回收率为103.31%,相对标准偏差为1.14%.结论 从植物形态、性状、显微特征方面可比较全面地对生药糙苏进行鉴别.高效液相色谱法简便、准确、重复性好,适用于糙苏药材的质量控制.Objective To establish a method for microscopic identification and determination of Shanzhiside methylester in Phlomis umbrosa Turcz and to provide the basis for the quality standards of phlomis umbrosa.Methods The microscope was used for pharmacognostic study on Phlomis umbrosa Turcz.The high performance liquid chromatography(HPLC) with a waters Sun FireTM C18 column(250 mm ×4.6 mm,5 μm) was used to measure the Shanzhiside methylester.The mobile phases was acetonitrile-water (9 ∶ 91) at 25 ℃;the detection wavelength was 236 nm;the flow rate was 1.0 ml/min and injection volume was 10 μl.Results Medicinal root and fine root of the Mongolian medicine Phlomis umbrosa Turcz were connected,presenting a unique shape;the transverse section of the root showed certain characteristics:large xylem conduit and obvious fissure.Powder-like reticulate vessels,varied size and shape and long fusiform shape of the wood fiber could be used as basic features of identifying the medicinal materials.The method in this study had a good linear relation within the range of 0.065 26-0.900 6 ng (r =0.999 7) for shanzhiside methylester.The average recovery rate of Shanzhiside methylester was 103.31% (relativestandarddeviation:1.14%).Conclusions From the aspects of the morphology,properties,microscopic characteristics,the raw drug of Phlomis umbrosa Turcz can be comprehensively identified.HPLC is a simple,accurate and reproducible method for quality control of Phlomis umbrosa Turcz.
