摘  要:目的探讨奥曲肽对博来霉素(BLM)诱导的大鼠肺纤维化的干预作用。方法 32只Wistar大鼠随机均分为四组,A、B、C三组以气管内滴注BLM 5mg/kg建立肺纤维化模型;D组给予等体积生理盐水作为空白对照。造模后,A组皮下注射奥曲肽50μg/kg,每天2次;B组腹腔注射地塞米松1mg/kg,每天1次;连续用药21d。第21天取材,采用HE及Masson染色法观察肺组织病理变化,碱水解法测肺组织羟脯氨酸(HYP)含量,ELISA法检测支气管肺泡灌洗液(BALF)中转化生长因子β1(TGF-β1)和结缔组织生长因子(CTGF)的水平。结果与D组比较,C组大鼠肺组织炎症和纤维化明显,肺组织HYP含量和BALF中TGF-β1和CTGF表达增高;而A、B两组的上述指标均低于C组(P<0.05)。结论奥曲肽能显著减轻肺纤维化大鼠肺组织肺泡炎及肺纤维化程度;其机制可能与其降低纤维化组织TGF-β1和CTGF表达有关。Objective To explore the effects of octreotide on bleomyein (BLM)-induced pulmonary fibrosis in rats. Methods A total of 32 Wistar rats was equally randomized into 4 groups. The rat models with pulmonary fibrosis were established by intra-traeheal injection of BLM 5 mg/kg in groups of A, B and C, after which the rats in group A were subcutaneously injected oetreotide 50μg/kg bid for 21 days and those in group B with intraperitoneal injection of dexamethasone 1mg/kg qd for 21 days. The rats in group D were taken as blank controls. Pathological changes of the lungs were observed by HE and Masson staining and hydroxyproline(HYP) in lung tissues was determined by alkaline hydrolysis. The expressions of TGF-β1 and CTGF in bronchoalveolar lavage fluid(BALF) were detected by ELISA. Results Compared to group D, the alveolitis and pulmonary fibrosis were obvious and the levels of HYP,TGF-β1 and CTGF in lung tissues were significantly increased in group C,which were lower in groups of A and B than those in group C(P〈0. 05). Conclusion Octreotide can ruduce the severity of alveolitis and pulmonary fibrosis induced by BLM in rats, which may be related to a decrease of the expressions of TGF-β1 and CTGF in lung tissues with fibrosis.
