摘　　要：为研究规模化猪场猪繁殖与呼吸综合征（PRRS）疫苗免疫猪群中PRRS病毒（PRRSV）感染及其变异情况,本研究采集了775份血清及21份病料样品进行病原检测分析,共分离获得了14株高致病性PRRSV（HP-PRRSV）分离株,分别将其命名为Henan-A1～Henan-A14.通过对其全基因组测序及比对分析结果显示,分离株与HP-PRRSV代表株JXA1、HuN4差异显著,同源性仅约为96％.此外,这14株分离株间差异较大,平均差异率达3.44％,呈现不同的变异方向.进一步研究表明,有5个分离株不能在Marc-145细胞中增殖,这是区别于其他HP-PRRSV的重要特征,表明部分分离株的细胞嗜性发生了改变.采用抗HP-PRRSV HuN4株血清对14株分离株进行中和试验,结果显示5份血清对其中9株分离株的中和活性较低,表明目前分离的大部分PRRSV分离株的中和抗原已经发生了较大的变异.本研究结果为有效预防PRRS提供了实验依据.In order to study the prevalance and genetic diversity of porcine reproductive and respiratory syndrome virus （PRRSV） in the vaccine immunized large-scale pig fxam, a total of 754 serum and 21 tissue samples of pigs were collected and subjuct to detection and isolation of PRRSV. Then, fourteen PRRSVs （named Henan-A1 to Henan-Al4） were isolated. By sequence homology analysis, we found that the 14 isolates not only have a low homology （ around 96%） with JXA1 or HUN4, but also shown difference among their genomes （mean difference is 3.44%）. In addition, 5 out of the 14 isolates failed to proliferate on MARC-145 cell, which was an important feature different from other HP-PRRSV, indicating that the cell tropism of the isolates had changed. Furthermore, the neutralizing antibody titer used to evaluate the 14 isolates in PAM cells with 5 neutralizing sera against HuN4 strain, respectively, and the result showed that 5 serum were barely able to neutralize 9 isolates, reflecting most of the PRRSVs currently isolated have a profound variation in neutralizing antigen.
