摘 要:目的研究C形臂X射线机扫描对小鼠的辐射损伤作用。方法随机数字表法将72只昆明雄性小鼠分成6组,设对照组、单次、两次、四次、八次、十六次扫描组。用C形臂X射线机全身扫描各组小鼠24 h后,总抗氧化能力检测试剂盒(ABTS快速法)、GSH检测试剂盒和活性氧检测试剂盒分别用于检测脾脏总抗氧化能力(T-AOC)、还原型谷胱甘肽(GSH)含量和活性氧(ROS)水平。流式细胞仪检测各照射组小鼠外周血淋巴和骨髓细胞的DNA损伤(γ-H2AX)水平(1 h后检测)和骨髓细胞周期进程变化(24 h后检测)。结果四次、八次和十六次C形臂X射线机扫描显著增加了脾脏中ROS的水平(P<0.05),十六次扫描极大的降低了GSH含量和T-AOC(P<0.05)。四次、八次和十六次C形臂X射线机扫描增加了小鼠外周血淋巴和骨髓细胞的γ-H2AX水平。十六次C形臂X射线机扫描增加了骨髓细胞G0/G1期和G2/M百分率(P<0.05),八次和十六次扫描降低S期百分率(P<0.05)。结论 C形臂X射线机扫描对小鼠产生了不同程度的辐射损伤。Objective To investigate radiation injuries induced by C- arm X ray scan in mice. Methods 72 healthy male kunming mice were randomly divided into control group,single,twice,four,eight and sixteen times of C- arm X ray scan.The mice were administered with different frequency of C- arm X ray scan; After 24 h,T- AOC,GSH and ROS were detected by T- AOC Assay Kit( ABTS method),GSH Assay Kit and ROS Assay Kit,respectively. The γ- H2 AX level in peripheral lymphocytes and bone marrow cells( cell cycle phase) were analyzed by flow cytometry. Results Compared with control,four,eight and sixteen times of scan increased ROS level,but eight and sixteen times of scan decreased GSH level,sixteen times of scan significantly decreased T- AOC levelin thyroid. Compared with control,four,eight and sixteen times of scan increased γ- H2 AX level in peripheral lymphocytes,eight and sixteen times of scan increased γ- H2 AX level in bone marrow cells. Compared with control,sixteen times of scan increased the percentages of G0/ G1 and G2/ M phases,while eight and sixteen times of scan decreased the percentages of S phases. Conclusion C- arm X ray scan produced different degrees of radiation injuries in mice.
