摘 要:目的观察艾塞那肽对高糖及低糖条件下氧化应激的干预情况及其在降低胰岛β细胞凋亡中的作用。方法体外培养大鼠胰腺β细胞株INS-1细胞,分为正常对照组(NC)、高糖组(HG)、低糖组DG和干预组即艾塞那肽高糖干预组(HE)、艾塞那肽低糖干预组(DE)、二甲双胍高糖干预组(HS)。相差显微镜观察细胞生长状态;流式细胞仪双染法及原位TUNEL法检测INS-1细胞凋亡情况;检测培养液中胰岛素(INS)、8-异前列腺素F 2α(8-iso-PGF2α)、丙二醛(MDA)、超氧化物歧化酶(SOD)水平。结果细胞凋亡指数及凋亡率HG、DG、HE、DE和HS组均高于NC组(P均<0.01或<0.05),HE组低于HG组(P均<0.05),DE组细胞凋亡指数低于DG组(P<0.05)。HG、DG、HE和DE组MDA及8-iso-PGF2α水平均高于NC组,SOD和INS水平低于NC组(P均<0.01或<0.05);HE组MDA、8-iso-PGF2α水平低于HG组(P<0.05),SOD、INS水平高于HG组(P<0.05);DE组MDA、8-iso-PGF2α水平低于DG组(P<0.05),SOD水平高于DG组(P<0.05);HE组INS水平高于DE组(P<0.05)。HS组INS水平低于HE组、MDA水平高于HE组(P<0.05)。结论艾塞那肽通过抑制包括高糖及低糖条件下所诱导的氧化应激水平,从而在一定程度上保护β细胞,减少细胞凋亡,有助于维持及提高β细胞数量。Objective To observe protective effects of Exenatide on the oxidative stress and apoptosis of rat pancreatic β cells under the condition of low Glucose and high Glucose. Methods The in vitro cultured INS - 1 cells were divided into six groups : the control group ( NC ) , high glucose group ( HG), low glucose group (DG), high glucose + Exenatide group(HE),low glucose group + Exenatide group (DG), high glucose + mefformin group(HS) , and these cells were cultured 48 hours respectively. The morphological change of cells were observed under the inverted microscope. The apoptosis rate of the INS - 1 cells was tested by the flow cytometry and TdT - mediated dUTP nick end labeling. The level of Insulin, 8 - different prostaglandins F_ (2 alpha), malondialdehyde ( MDA), superoxide dismutase (SOD) were evaluated in cell culture fluid. Results In the group of HG, DG, HE, DE and HS, the apoptosis rates and apoptosis indexs of the INS - 1 cells were higher than those of the control group ( P 〈 0.01 or 〈 0.05 ). The levels of the indexs and rates in HE group were lower than those in HG group. Apoptosis indexs in ED group was lower than that in DG group. Compared with NC group, in HG, DG, HE and DE group the levels of MDA and 8 - iso - PGF2αcontent increased significantly while SOD expression and the levels of Insul decreased ob-viously ( P 〈 0.01 or 〈 0.05 ). After the Exenatide intervention, oxidative stress levels were significantly inhibi-ted. The levels of MDA and 8 - iso - PGF2αin HE and DE group were lower than those in HG and DG group respectively(P 〈 0.05). SOD expression levels in HE and DE group were higher than those in HG and DG group ( P 〈 0.05 ). The levels of Insul in DE and HS group were lower than those in HE ( P 〈 0.05 ). The lev-els of MDA in HS were higher than those in HE group (P 〈 0.05 ).
